HSP70、JNK和p38在放線菌素D誘導(dǎo)的A549細胞凋亡中的作用

【摘要】:目的:探討HSP70、JNK和p38在放線菌素D(Act D)誘導(dǎo)的人肺腺癌A549細胞凋亡中的作用。方法:采用MTT法確定Act D對A549細胞的染毒劑量。將對數(shù)生長期的A549細胞分為DMSO對照組、Act D染毒組、熱處理(42℃,30 min)+Act D組、JNK抑制劑SP600125+Act D組和p38 MAPK抑制劑SB203580+Act D組進行處理。采用流式細胞術(shù)檢測各組細胞的凋亡率,Western blot檢測各組細胞中HSP70、p-JNK、p-p38和Caspase-3的表達水平。結(jié)果:熱處理、SP600125和SB203580均能降低Act D誘導(dǎo)的A549細胞的凋亡率(F=7 904.576,P0.001)。熱處理+Act D組的HSP70表達水平高于其余各組(F=46.640,P0.001),而p-JNK、p-p38和Caspase-3的表達水平均低于Act D處理組(F=122.381、44.104和50.650,P0.05)。Act D能提高Caspase-3的表達水平,且熱處理、SP600125和SB203580均能降低Act D引起的Caspase-3的高表達狀態(tài)。結(jié)論:熱處理誘導(dǎo)產(chǎn)生的HSP70、JNK通路抑制劑SP600125和p38 MAPK通路抑制劑SB203580均可以抑制由Act D誘導(dǎo)的A549細胞凋亡。
