以EGFR為靶點(diǎn)siRNA的初步研究

【摘要】: 表皮生長(zhǎng)因子受體(epidermal growth factor receptor, EGFR)的過度表達(dá)被認(rèn)為是腫瘤細(xì)胞過度增殖的重要因素之一。抑制EGFR的過度表達(dá)可以有效抑制細(xì)胞的過度增殖,從而達(dá)到腫瘤臨床治療的目的。RNA干擾(RNA interference, RNAi)技術(shù)可以高效高特異性地抑制特異蛋白的表達(dá)量,在腫瘤的治療方面具有良好的應(yīng)用前景。 本研究在體外培養(yǎng)角質(zhì)形成細(xì)胞和舌鱗癌細(xì)胞的基礎(chǔ)上,根據(jù)小干擾RNA(small interfering RNA, siRNA)的設(shè)計(jì)原則設(shè)計(jì)并合成以EGFR為靶點(diǎn)的siRNA,通過RNAi技術(shù)研究其對(duì)角質(zhì)形成細(xì)胞和舌鱗癌細(xì)胞EGFR mRNA表達(dá)的抑制作用。從實(shí)驗(yàn)結(jié)果中篩選出一條高效的以EGFR為靶點(diǎn)siRNA。研究其對(duì)舌鱗癌細(xì)胞增殖的抑制作用。該研究為以EGFR為靶點(diǎn)的siRNA臨床治療鱗癌、銀屑病等EGFR過度表達(dá)的臨床疾病研究提供一種新的實(shí)驗(yàn)方法,為以RGFR為靶點(diǎn)藥物在臨床疾病治療中的應(yīng)用提供一定的實(shí)驗(yàn)依據(jù)。
[Abstract]:The overexpression of epidermal growth factor receptor (epidermal growth factor receptor, EGFR) is considered to be one of the important factors in the proliferation of tumor cells. Inhibiting the overexpression of EGFR can effectively inhibit the excessive proliferation of cells and achieve the purpose of clinical treatment of tumor. RNA interference (RNA interference, RNAi) technique can inhibit the expression of specific protein efficiently and specifically. It has a good prospect in the treatment of tumor. In this study, keratinocytes and tongue squamous carcinoma cells were cultured in vitro, and siRNA, targeting EGFR was designed and synthesized according to the design principle of small interference RNA (small interfering RNA, siRNA). The inhibition of EGFR mRNA expression in keratinocytes and tongue squamous carcinoma cells was studied by RNAi technique. A highly efficient siRNA. targeting EGFR was selected from the experimental results. To study its inhibitory effect on the proliferation of tongue squamous cell carcinoma cells. This study provides a new experimental method for the clinical treatment of squamous cell carcinoma, psoriasis and other overexpressed EGFR diseases with siRNA targeted at EGFR, and provides a certain experimental basis for the application of RGFR as a target drug in the treatment of clinical diseases.
