microRNA-125a-5p對巨噬細胞THP-1泡沫化過程的影響及其機制

【摘要】:目的探討microRNA-125a-5p(miR-125a-5p)對巨噬細胞THP-1泡沫化過程的影響及其機制。方法在Lipao2000的介導(dǎo)下,將miR-125a-5p的增強劑(mimics)/抑制劑(inhibition)(終濃度均為50 nmol/L)分別轉(zhuǎn)染THP-1細胞24 h后,加入終濃度為100 mg/L的低密度脂蛋白(oxygenized-low density lipoprotein,ox-LDL),繼續(xù)培養(yǎng)48 h,同時設(shè)空白對照組(未轉(zhuǎn)染)及ox-LDL組(僅加入終濃度為100 mg/L的ox-LDL)。采用油紅O染色和脂質(zhì)染色的半定量分析法檢測miR-125a-5p對THP-1細胞泡沫化的影響;Cholesterol/Cholesteryl Ester Quantitation Kit檢測miR-125a-5p對THP-1細胞泡沫化過程中脂質(zhì)蓄積的影響;Real-time PCR和Western blot法檢測miR-125a-5p對THP-1細胞中;o酶A膽固醇;D(zhuǎn)移酶-1(acyl-coenzyme A:cholesterol acyltrasferase-1,ACAT-1)、ATP結(jié)合盒轉(zhuǎn)運體A1(ATP binding casstte transporter A1,ABCA1)及細胞清道夫受體-A1(scavenger receptor-A1,SR-A1)mRNA及蛋白表達水平的影響。結(jié)果與ox-LDL組比較,ox-LDL+mimics組巨噬細胞泡沫化程度及脂質(zhì)蓄積量均明顯下降(P均0.05),ox-LDL+inhibition組均明顯增加(P均0.05);ox-LDL+mimics組ACAT-1和SR-A1 mRNA及蛋白表達水平明顯降低(P均0.05),而ABCA1 mRNA及蛋白表達水平明顯升高(P均0.05);ox-LDL+inhibition組ACAT-1和SR-A1 mRNA及蛋白表達水平明顯增加(P均0.05),而ABCA1 mRNA及蛋白表達水平明顯降低(P均0.05)。結(jié)論 miR-125a-5p可通過調(diào)解ACAT-1、ABCA1和SR-A1的表達水平,減少巨噬細胞內(nèi)脂質(zhì)類的堆積,抑制巨噬細胞向泡沫細胞的轉(zhuǎn)化,從而發(fā)揮抗動脈粥樣硬化(atheroscler-osis,As)的作用。
[Abstract]:Objective to investigate the effect and mechanism of microRNA-125a-5p (miR-125a-5p) on THP-1 foaming of macrophages. Methods THP-1 cells were transfected with miR-125a-5p enhancer (mimics) / (inhibition) (50 nmol/L) mediated by Lipao2000 for 24 h. Oxygenized-low density lipoprotein, with final concentration of 100 mg/L was added. Conclusion miR-125a-5p can mediate the expression of ACAT-1,ABCA1 and SR-A1, reduce the accumulation of lipids in macrophages, inhibit the transformation of macrophages to foam cells, and thus play a role in anti-atherosclerosis (atheroscler-osis,).
