聲誘發(fā)短潛伏期負電位(ASNR)的豚鼠模型

【摘要】: 目的:ASNR為強聲刺激出現(xiàn)的前庭源性電位,重度感音性聾及球囊功能正常耳為其出現(xiàn)的必備條件。本研究通過驗證豚鼠ASNR與VEMP出現(xiàn)的一致性來建立ASNR豚鼠模型,即建立重度感音性聾且球囊功能正常的豚鼠,用短聲刺激誘發(fā)ASNR。 方法:將27只健康豚鼠隨機分為兩組,正常對照組16只(32耳)、給藥致聾組11只(22耳)。致聾組經(jīng)給藥(400mg/kg Ka皮下注射,一小時后,40mg/kg Ea頸靜脈注射)致聾后至少恢復一周。對所有動物進行聽覺前庭功能檢查包括(ABe測試、VEMP測試及冰水試驗)。致聾組豚鼠根據(jù)ASNR的出現(xiàn)分為兩組(ASNR組、非ASNR組)。 結果:11只豚鼠(22耳)中8耳出現(xiàn)ASNR,出現(xiàn)率為36.4%,其閾值范圍為120-130dB SPL(平均124.4±4.96dB SPL),潛伏期(1.75ms-2.60ms),平均為2.15±0.27ms,閾值平均潛伏期2.34±0.18ms。ASNR組8耳皆出現(xiàn)VEMP,VEMP閾值及正負峰潛伏期與正常組比較均無統(tǒng)計學差異,未出現(xiàn)VEMP的10耳,皆無ASNR,非ASNR組中有四耳出現(xiàn)VEMP,其閾值及正負峰潛伏期與正常組比較亦無統(tǒng)計學差異。致聾組VEMP、ASNR的引出與冰水誘發(fā)正常眼震均無相關關系。 結論:實驗應用前庭球囊測試指標(VEMP)驗證了ASNR的出現(xiàn)與其具有一致性,從而證明了ASNR與VEMP同起源于球囊,而與冰水試驗結果無關。本實驗在重度感音性聾且球囊功能正常的豚鼠身上誘發(fā)出ASNR,通過耳毒藥物法首次成功建立ASNR動物模型。
[Abstract]:Objective: ASNR is the vestibular potential of strong sound stimulation, and severe sensorineural hearing loss and normal balloon function are the necessary conditions. The aim of this study was to establish a guinea pig model of ASNR by verifying the consistency between ASNR and VEMP, that is, to establish a guinea pig with severe sensorineural hearing loss and normal balloon function. ASNR. was induced by short sound stimulation. Methods: 27 healthy guinea pigs were randomly divided into two groups: the control group (n = 16) (n = 16) and the drug induced deafness group (n = 11) (n = 22). The deafness group recovered at least one week after administration of 400mg/kg Ka (subcutaneous injection, 1 hour later, 40mg/kg Ea via jugular vein). Auditory vestibular tests were performed on all animals (ABe test, VEMP test and ice water test). According to the appearance of ASNR, guinea pigs in deafness group were divided into two groups (ASNR group, non-ASNR group). Results: in 11 guinea pigs (22 ears), the occurrence rate of ASNR, in 8 ears was 36.4, and the threshold value was 120-130dB SPL (mean 124.4 鹵4.96dB SPL), latency (1.75ms-2.60ms), average 2.15 鹵0.27 Ms). There was no significant difference in VEMP,VEMP threshold and positive and negative peak latency in 8 ears of the group with 2. 34 鹵0.18ms.ASNR, 10 ears without VEMP, and 4 ears with no ASNR, in non ASNR group. There was no significant difference in the threshold and latency of positive and negative peaks between the two groups. There was no correlation between VEMP,ASNR elicitation and normal nystagmus induced by ice water in deafness group. Conclusion: the vestibular balloon test index (VEMP) was used to verify the consistency between the appearance of ASNR and ASNR, which proved that both ASNR and VEMP originated from the balloon, but had nothing to do with the results of ice water test. In this study, ASNR, was successfully established in guinea pigs with severe sensorineural deafness and normal balloon function by ototoxic method.
