【摘要】：目的研究鎘對(duì)成年雄性大鼠脊髓下腰段SNB和DLN運(yùn)動(dòng)神經(jīng)元超微結(jié)構(gòu)和雄激素受體(AR)表達(dá)的影響。方法成年雄性大鼠腹腔內(nèi)反復(fù)小劑量注射氯化鎘(0.5mg/kg體重和1.0mg/kg體重交替注射)及丙酸睪酮(TP)500μg皮下注射后,采血測(cè)定血清睪酮含量,取脊髓L5-6節(jié)段進(jìn)行SNB和DLN運(yùn)動(dòng)神經(jīng)元AR免疫組化及光密度(OD)分析,透射電鏡觀察SNB和DLN運(yùn)動(dòng)神經(jīng)元超微結(jié)構(gòu),采用原子吸收石墨爐法測(cè)定脊髓鎘含量。結(jié)果染鎘后脊髓內(nèi)鎘含量升高,血清睪酮水平在染鎘30d時(shí)明顯下降。染鎘7d、10d、15d,神經(jīng)元核AR免疫染色平均OD值降低,TP皮下注射2h,AR平均OD值無明顯升高;染鎘30d,AR平均OD值進(jìn)一步降低,TP皮下注射2h,AR平均OD值增加,但是只達(dá)到單純?nèi)炬k7d、10d、15d時(shí)水平。染鎘后,神經(jīng)元超微結(jié)構(gòu)無明顯改變。結(jié)論鎘可以蓄積在成年大鼠脊髓內(nèi),導(dǎo)致SNB和DLN運(yùn)動(dòng)神經(jīng)元功能損傷,可能與鎘所致性功能障礙有關(guān)。
[Abstract]:Objective to study the effects of cadmium on the ultrastructure of SNB and DLN motor neurons and the expression of androgen receptor (AR) in the lower lumbar spinal cord of adult male rats. Methods male adult rats were injected with cadmium chloride (0.5mg/kg body weight and 1.0mg/kg body weight alternately) and testosterone propionate (TP) 500 渭 g) subcutaneously injected intraperitoneally. Serum testosterone levels were measured. The spinal cord L5-6 segments were taken for AR immunohistochemistry and optical density (OD) analysis of SNB and DLN motor neurons. The ultrastructure of SNB and DLN motor neurons was observed by transmission electron microscope. The content of CD in spinal cord was measured by atomic absorption graphite furnace method. Results the content of CD in spinal cord increased after cadmium exposure, and the serum testosterone level decreased significantly after 30 days of cadmium exposure. The mean OD value of AR immunostaining in neurons decreased after 7 days and 10 days and 15 days after exposure to cadmium, but the average OD value of AR was not significantly increased at 2 hours after subcutaneous injection of TP. After 30 days of cadmium exposure, the average OD value of AR decreased further, and the average OD value of TP increased at 2 h after subcutaneous injection, but only reached the level of 10 days or 15 days after exposure to cadmium alone for 7 days or 10 days. The ultrastructure of neurons did not change after cadmium exposure. Conclusion cadmium accumulates in the spinal cord of adult rats and results in functional damage of SNB and DLN motor neurons, which may be related to the sexual dysfunction caused by cadmium.
