河北盧龍縣豬與人G3型A組輪狀病毒進化關系研究
[Abstract]:The main gene fragments VP7,VP4,VP6,VP2 and NSP2-5 of GARV LLZ212 of Lulong G3 piglets were amplified and their genotypes were determined by amplification of VP7,VP4,VP6,VP2 and NSP2-5 in the virus diarrhea laboratory of the Central Center for Disease Control and Prevention of China. The evolutionary relationship between G3 GARV and local people was also discussed. Reverse transcription-polymerase chain reaction (Reverse transcription-polymerase chain reaction,RT-PCR) was used to amplify VP7,VP4,VP6,VP2 and NSP2-5 from LLZ212 samples and G3 GARV positive samples from 6 local children. Group A rotavirus automatic typing tool (RotaC v2.0) was used to type each fragment. The homology and phylogenetic analysis of reference strains on GenBank were carried out by using DNAStar and MEGA5.0 software packages. The LLZ212 genotypes of Lulong pigs were G3-P [8] -I5-C1-N1-T1-E1-H1G, all of which were G3-P [8]-I1-C1-N1-T1-E1-H1.LLZ212 gene fragment VP7,VP4,NSP4 and NSP5, respectively. Wa and RMC321 have the highest homology, and the evolutionary tree is in the same sub-branch. The VP6,NSP2 and NSP3 fragments of LLZ212 gene were most homologous to GARV PRG921 strain, and VP2 gene fragment was the most homologous to GARV OSU strain, and the phylogenetic tree was in the same subbranch. It was confirmed that a rare G3-P [8]-I5-C1-N1-T1-E1-H1 porcine GARV, might be a human-to-pig rematch strain. The LLZ212 gene fragments VP7,VP4,NSP4 and NSP5 share the highest homology with human GARV, and LLZ212 gene fragment VP6,VP2,. NSP2 and NSP3 porcine GARV have the highest homology.
