大鼠機(jī)械性腦損傷蛋白質(zhì)表達(dá)譜的變化及意義

【摘要】:目的:研究閉合性顱腦損傷大鼠血清和海馬蛋白質(zhì)表達(dá)譜,探尋腦損傷生物標(biāo)志蛋白。 方法:選雄性SD 大鼠隨機(jī)分為手術(shù)對(duì)照組及損傷后4 h、8 h、12 h、24 h 和48 h 組,復(fù)制Marmarou’s 落體打擊大鼠腦損傷模型。分別進(jìn)行神經(jīng)行為學(xué)評(píng)價(jià)、常規(guī)HE 染色、尼氏染色和酯化銀染色;并用弱陽(yáng)離子交換芯片(WCX2)和金屬親和吸附芯片(IMAC3)結(jié)合表面增強(qiáng)激光解析電離飛行時(shí)間質(zhì)譜技術(shù)分析大鼠閉合性腦損傷后不同時(shí)間血清和海馬中蛋白質(zhì)表達(dá)譜的改變。 結(jié)果:1. 臨床表現(xiàn):損傷組大鼠于打擊后有84 只大鼠(70%)出現(xiàn)抽搐同時(shí)有呼吸暫停,持續(xù)約15s;在沒(méi)有出現(xiàn)抽搐的大鼠中有11 只大鼠(9.17%)出現(xiàn)驚厥,持續(xù)約30s;17 只大鼠(14.3%)傷后有肢體活動(dòng)受限,表現(xiàn)為單肢或單側(cè)肢體活動(dòng)受限。12 只大鼠(10%)于打擊后出現(xiàn)持續(xù)呼吸抑制和全身抽搐,幾十秒鐘后呼吸停止,死亡。 2. 神經(jīng)行為學(xué)評(píng)價(jià):大鼠腦損傷后平衡能力、翻身、支頭、行走、逃避和四肢功能均較損傷前有所減弱,評(píng)分降低。腦損傷后1 h、12 h 大鼠行為學(xué)評(píng)分與損傷前相比有顯著性差異(P0.01),損傷后24 h、48 h 與損傷前比較無(wú)顯著性差異。 3. 大體病理:損傷組大鼠可見(jiàn)蛛網(wǎng)膜下腔出血,分布廣泛,無(wú)明顯大面積挫傷或局灶性挫裂傷;切面可見(jiàn)胼胝體點(diǎn)狀出
[Abstract]:Aim: to study the protein expression profiles of serum and hippocampus in rats with closed brain injury and to explore the biomarkers of brain injury. Methods: male SD rats were randomly divided into two groups: the control group and the control group. The rats were randomly divided into two groups: the control group and the 24 h and 48 h groups after 4 h, 8 h and 12 h after injury. The model of brain injury was induced by Marmarou's falling body. Neurobehavioral evaluation, routine HE staining, Nissl staining and esterification silver staining were performed. The changes of protein expression profiles in rat serum and hippocampus at different times after closed brain injury were analyzed by weakly cation exchange chip (WCX2) and metal affinity adsorption chip (IMAC3) combined with surface enhanced laser desorption ionization time of flight mass spectrometry. The result is 1: 1. Clinical manifestations: 84 rats (70%) in the injury group had convulsions and apnea for about 15 s, 11 rats (9.17%) showed convulsion for about 30 s. 17 rats (14.3%) had limited limb movement after injury, which was characterized by limited movement of one or one limb, 12 rats (10%) had sustained respiratory inhibition and systemic convulsions after striking, and after dozens of seconds, breathing stopped and died. 2. Neurobehavioral evaluation: after brain injury, the ability of balance, turning over, branching, walking, escaping and limb function were all weakened and the score was lower. There was a significant difference in behavioral score between 1 hour and 12 h after brain injury (P0.01), but there was no significant difference between 24 h and 48 h after injury. 3. Gross pathology: subarachnoid hemorrhage was found in the injury group, with no obvious large area contusion or focal contusion, and corpus callosum punctate on the section.
