細胞凋亡抑制因子survivin在精子發(fā)生過程中的作用研究

【摘要】: 目的:檢測細胞凋亡抑制因子survivin mRNA及survivin蛋白在精索靜脈曲張大鼠睪丸組織中的表達,初步探討其在精子發(fā)生過程中的意義。 方法:1.利用Saypol法部分結扎大鼠左腎靜脈,復制精索靜脈曲張大鼠模型,建立生精障礙的動物模型。2.采用逆轉錄聚合酶鏈反應(RT-PCR)法及鏈霉菌抗生物素-過氧化酶連接(SP)免疫組織化學方法檢測:(1)精索靜脈曲張大鼠睪丸組織中survivin mRNA的表達。(2)精索靜脈曲張大鼠睪丸組織中survivin蛋白的表達。 結果:1.精索靜脈曲張影響大鼠精子的發(fā)生,隨著曲張時間的延長,其睪丸損害越重P0.05㖞。2. survivin mRNA及survivin蛋白在正常大鼠睪丸組織中有表達。3. survivin mRNA和survivin蛋白的表達水平與精索靜脈曲張的時間有關,曲張的時間越長,survivin在大鼠睪丸組織中的表達越少P0.05㖞。免疫組化顯示:survivin蛋白主要表達于次級精母細胞和精子細胞的胞漿,在睪丸間質細胞中有少量表達。4.survivin的表達與睪丸組織的病理分級有關,在減數(shù)分裂前出現(xiàn)成熟障礙的睪丸組織中沒有表達,而減數(shù)分裂后發(fā)生成熟障礙的標本的表達量與正常睪丸沒有區(qū)別。 結論:1.精索靜脈曲張是引起精子發(fā)生異常的原因之一,與細胞凋亡抑制因子survivin表達密切相關。2. survivin的表達與精子發(fā)生早期維持細胞的增殖有關。它可能是精子發(fā)生中的一個關鍵因子。
[Abstract]:Aim: to detect the expression of apoptosis inhibitor survivin mRNA and survivin protein in testis of varicocele rats and to explore their significance in spermatogenesis. Methods: 1. The rat model of varicocele was established by partial ligation of left renal vein by Saypol method. 2. Reverse transcriptase polymerase chain reaction (RT-PCR) and streptomyces biotin-peroxidase (SP) were used to detect the expression of survivin mRNA in testis of varicocele rats. Expression of survivin protein in testis of varicocele rats. The result is 1: 1. Varicocele affects spermatogenesis in rats. With the extension of varicocele, the more serious the testicular damage is? P0.05? 2. Survivin mRNA and survivin protein were expressed in normal rat testis. 3. 3. The expression of survivin mRNA and survivin protein was related to the time of varicocele. The longer the varicocele was, the less survivin was expressed in rat testis. Immunohistochemical staining showed that survivin protein was mainly expressed in the cytoplasm of secondary spermatocytes and spermatocytes, and a little in stromal cells of testis. The expression of 4.survivin was related to the pathological grade of testis. There was no expression in testis with maturation disorder before meiosis, but the expression of maturation disorder after meiosis was not different from that in normal testis. Conclusion: 1. Varicocele is one of the causes of abnormal spermatogenesis, which is closely related to the expression of apoptosis inhibitor survivin. 2. The expression of survivin is related to the proliferation of early spermatogenesis maintenance cells. It may be a key factor in spermatogenesis.
