單克隆抗體電荷異構(gòu)體分離方法優(yōu)化

【摘要】:正單克隆抗體是復(fù)雜的四聚體糖蛋白,常呈現(xiàn)微觀不均一性,即"異質(zhì)性",包括電荷、疏水、形態(tài)等相關(guān)的異構(gòu)體[1]。這些異構(gòu)體可能來(lái)自于抗體分子復(fù)雜的生物合成途徑,如細(xì)胞系及培養(yǎng)工藝[2],也可能來(lái)自于純化、制劑等制造過(guò)程以及貯存過(guò)程的任何階段[1]。其中由于抗體分子所帶電荷差異造成的異質(zhì)性稱(chēng)為電荷異構(gòu)體,一般分為酸性異構(gòu)體和堿性異構(gòu)體,產(chǎn)生原因主要與翻譯后修飾有關(guān)。電荷異
[Abstract]:Positive monoclonal antibodies are complex tetramer glycoproteins, which often exhibit microscopic heterogeneity, including charge, hydrophobicity, morphology and other related isomers [1]. These isomers may be derived from complex biosynthetic pathways of antibody molecules, such as cell lines and culture processes [2], or from any stage in the manufacturing process, such as purification, preparation, and storage [1].
