高糖誘導(dǎo)的星形膠質(zhì)細(xì)胞凋亡的氧化應(yīng)激通路及總?cè)岬母深A(yù)作用

【摘要】:目的研究總?cè)釋Ω咛钦T導(dǎo)的星形膠質(zhì)細(xì)胞凋亡的干預(yù)作用及機(jī)制。方法采用體外分離乳鼠星形膠質(zhì)細(xì)胞培養(yǎng)并分為對照組、模型組、總?cè)岣深A(yù)組。高糖溫孵后提取細(xì)胞mRNA和蛋白質(zhì)分析細(xì)胞凋亡蛋白Bax/Bcl2及NADPH氧化酶亞基p22的表達(dá)。結(jié)果模型組細(xì)胞Bax表達(dá)上調(diào)而Bcl2表達(dá)下調(diào),同時p22表達(dá)上調(diào)。總?cè)扑岣深A(yù)后上述蛋白表達(dá)部分得到恢復(fù)。結(jié)論總?cè)峥赏ㄟ^抑制p22上調(diào)抑制高糖引起的星形膠質(zhì)細(xì)胞凋亡,減輕細(xì)胞的損傷。
[Abstract]:Objective to study the effect and mechanism of total tricitric acid on apoptosis of astrocytes induced by high glucose. Methods astrocytes were isolated from neonatal rats in vitro and divided into three groups: control group, model group and total tricitric acid intervention group. MRNA and protein expression of apoptotic protein Bax/Bcl2 and NADPH oxidase subunit p22 were analyzed. Results in the model group, the expression of Bax was up-regulated, the expression of Bcl2 was down-regulated, and the expression of p22 was up-regulated. After total triterpene acid intervention, the above protein expression was partially recovered. Conclusion TSA can inhibit the apoptosis of astrocytes induced by high glucose by inhibiting the up-regulation of p22 and attenuating the injury of astrocytes.
