【摘要】：目的:探討體外分離培養(yǎng)的新生兔氣管軟骨細胞的生物學特性。方法:通過酶消化法體外分離培養(yǎng)新生兔氣管軟骨細胞;倒置顯微鏡觀察軟骨細胞形態(tài)及生長狀況;電鏡觀察軟骨細胞超微結構;運用real-time PCR、免疫細胞化學染色和甲苯胺藍染色檢測氣管軟骨細胞分泌的細胞外基質成分。結果:體外分離、培養(yǎng)的兔氣管軟骨細胞呈短小三角形或不規(guī)則形貼壁生長。超微結構顯示細胞較多突起,孔隙較多,胞質豐富,細胞器發(fā)達,細胞內可見大量蛋白分泌物。軟骨細胞表達I、II型膠原、蛋白聚糖等,以II型膠原和蛋白聚糖表達為主。免疫細胞化學染色II型膠原和SOX9陽性,I型膠原弱陽性。甲苯胺藍染色陽性。結論:適宜的酶消化單層培養(yǎng)法獲得的新生兔氣管軟骨細胞具有分泌軟骨細胞外基質成分的特性,可初步為體外構建組織工程氣管治療新生兔氣管狹窄的實驗研究提供種子細胞。
[Abstract]:Objective: to investigate the biological characteristics of trachea chondrocytes isolated from neonatal rabbits in vitro. Methods: neonatal rabbit trachea chondrocytes were isolated and cultured by enzyme digestion in vitro, the morphology and growth of chondrocytes were observed by inverted microscope, the ultrastructure of chondrocytes were observed by electron microscope. The extracellular matrix secreted by tracheal chondrocytes was detected by real-time PCR, immunocytochemical staining and toluidine blue staining. Results: cultured rabbit trachea chondrocytes grew in short triangle or irregular shape. The ultrastructure showed that the cells had more protuberances, more pores, abundant cytoplasm, well developed organelles, and a large number of protein secretions could be seen in the cells. Chondrocytes expressed type II collagen and proteoglycan, mainly II type collagen and proteoglycan. Immunocytochemical staining showed that II type collagen and SOX9 were positive, and type I collagen was weakly positive. Toluidine blue staining was positive. Conclusion: the new rabbit tracheal chondrocytes obtained by enzyme digestion monolayer culture have the characteristic of secreting extracellular matrix of chondrocytes. It may provide seed cells for the experimental study of trachea stenosis in newborn rabbits with tissue engineering trachea in vitro.
