人類胚胎肺不同時期SP-A和SP-D的表達及其意義

【摘要】:肺表面活性物質(zhì)相關(guān)蛋白A(pulmonary surfactant associated protein A, SP-A)和表面活性物質(zhì)相關(guān)蛋白D(pulmonary surfactant associated protein D, SP-D)主要分布于支氣管表面和肺泡氣液界面上,主要由肺泡Ⅱ型細胞(alveolar epithelia cell Ⅱ, ACE Ⅱ)所分泌,屬膠凝素家族的重要成員,F(xiàn)已證實,SP-A、SP-D是肺部重要的天然免疫防御分子,不僅能清除病原體,還參與免疫、炎癥及過敏反應(yīng)的調(diào)節(jié),作為判斷胎肺發(fā)育成熟度以及診斷某些肺部疾病的臨床指標。 本研究旨在觀察人類胚胎肺組織發(fā)育的一般規(guī)律,分析不同時間點胚肺SP-A、SP-D蛋白合成和分泌的情況,初步探討胎肺成熟度與SP-A、SP-D生成的關(guān)系。并根據(jù)胚肺發(fā)育中這些免疫防御調(diào)節(jié)性因子(SP-A、SP-D)表達的情況,闡明早產(chǎn)兒或新生兒肺部感染及其局部免疫防御的生理病理學基礎(chǔ),為SP-A和SP-D替代療法提供理論依據(jù)。 材料和方法 收集水囊引產(chǎn)的妊娠10周-36周正常胎兒44例及正常新生兒5例和正常成人2例,取其肺組織固定、包埋制成石蠟切片,HE染色觀察肺組織各個發(fā)育階段的成熟度,免疫組化檢測SP-A、SP-D分子時相性表達特征。
