白念珠菌雙形態(tài)性的影響因素的研究

【摘要】: 目的: 探討白念珠菌雙形態(tài)性形成的相關(guān)影響因素,尋找控制菌絲形成的關(guān)鍵基因。 方法: 1、分離培養(yǎng)八株不同來源的白念珠菌(標準菌株和臨床標本分離菌株),控制白念珠菌菌絲生長的影響因素,包括孢子濃度、培養(yǎng)溫度、培養(yǎng)基成分和pH值,觀察菌絲的生長特點和形成率的變化。 2、分別采用-80℃研磨、液氮研磨、酸洗玻璃珠三種破壁方法破壁,用TRIzol試劑提取白念珠菌菌絲相和酵母相的總RNA,對提取的RNA進行定性定量及完整性分析。 3、通過設(shè)置內(nèi)參的半定量逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)的方法測定白念珠菌雙形態(tài)性的四個相關(guān)調(diào)控基因EFG1、HWP1、HYR1、HGC1的mRNA在菌絲相和酵母相中的表達差異。 4、將八株白念珠菌分別接種在顯色培養(yǎng)基和玉米吐溫培養(yǎng)基上,用NB-UVB臨床照射劑量(0.4-3.0J/cm~2)對其進行3次/天,共7天的照射,觀察菌絲和酵母的生長及形態(tài)變化。 結(jié)果: 1、菌絲具有聚集生長成絮狀物的特點。在pH=5.0時,菌絲上的孢子易脫落;在pH=9.0時,易形成較大分生孢子;中性pH值最有利于純菌絲的生長。孢子濃度5×10~6/ml,在含10%胎牛血清pH7.0的PRMI1640培養(yǎng)基中,38℃靜置培養(yǎng)4到8小時,菌絲的形成效果好。菌絲純度大于95%。 2、三種破壁方法提取白念珠菌酵母相RNA的濃度(x±s,ug/ul)分別為:-80℃研磨法(0.464±0.101,ug/ul)、酸洗玻璃珠法(0.911±0.413,ug/ul)、液氮研磨法(1.418±0.524,ug/ul)。而且后兩者RNA的完整性比前者好。 3、HWP1和HGC1基因在菌絲相有表達,而在酵母相無表達。EFG1和HYR1基因在菌絲相和酵母相的表達沒有明顯差異。 4、隨著UVB照射劑量的增加,菌絲形成逐漸減少,菌絲上生長的分生孢子逐漸增多。照射劑量超過1.5J/cm~2念珠菌的生長受到明顯抑制。 結(jié)論: 1、血清、中性pH值、適當溫度有利于菌絲生長。 2、液氮研磨破壁和酸洗玻璃珠振蕩破壁為較好的破壁提取白念珠菌RNA的方法,其中液氮研磨法最好。 3、NB-UVB照射劑量在0.4—3.0J/cm~2的時候,,可抑制菌絲生長。 4、HWP1和HGC1基因為菌絲特異表達基因。
