【摘要】：目的 研究D15S659和D9S925基因座的群體遺傳學(xué)和等位基因序列特征，積累群體遺傳學(xué)數(shù)據(jù)，，探討其法醫(yī)學(xué)應(yīng)用價值，為法醫(yī)遺傳學(xué)提供新的候選遺傳標(biāo)記。 方法 以109名華東地區(qū)漢族無血緣關(guān)系個體的靜脈血為研究樣本。用Chelex法提取DNA，對D15S659和D9S925基因座進(jìn)行PCR擴(kuò)增；非變性聚丙烯酰胺凝膠連續(xù)緩沖系統(tǒng)垂直電泳進(jìn)行分型。利用Modified-Powerstats標(biāo)準(zhǔn)版軟件對分型結(jié)果進(jìn)行統(tǒng)計(jì)分析，獲得各基因座的等位基因頻率、基因型計(jì)數(shù)及法醫(yī)群體遺傳學(xué)數(shù)據(jù)。 結(jié)果 D15S659和D9S925基因座在華東漢族群體中等位基因數(shù)分別為8和6；雜合度分別為0.826和0.798；個人識別率分別為0.944和0.894；非父排除率分別為0.648和0.596。 結(jié)論 在我國華東地區(qū)漢族人群中，D15S659和D9S925基因座等位基因頻率分布較好，提供的多態(tài)信息量大。具有較高的非父排除率和個人識別能力，適合于法醫(yī)學(xué)應(yīng)用，是群體遺傳學(xué)研究和法科學(xué)應(yīng)用較理想的候選STR基因座。
[Abstract]:Objective to study the population genetics and allelic sequence characteristics of D15S659 and D9S925 loci, accumulate population genetic data, and explore the value of forensic application to provide new candidate genetic markers for forensic genetics. Methods the venous blood samples were collected from 109 unrelated individuals of Han nationality in East China. The D15S659 and D9S925 loci were amplified by PCR with DNA, extracted by Chelex method, and typed by vertical electrophoresis of nondenatured polyacrylamide gel continuous buffer system. The allele frequency, genotype count and forensic population genetic data of each locus were obtained by statistical analysis of typing results with Modified-Powerstats standard software. Results the alleles of D15S659 and D9S925 loci were 8 and 6, heterozygosity were 0.826 and 0.798, personal identification rates were 0.944 and 0.894, non-paternal exclusion rates were 0.648 and 0.596, respectively. Conclusion the allelic frequencies of D15S659 and D9S925 loci are well distributed and the amount of polymorphic information is large in the Han population in East China. It has high non-paternal exclusion rate and personal recognition ability and is suitable for forensic application. It is an ideal candidate STR locus for population genetics and forensic science.
