人胎咽、食管肌層中a-SMA、MHC-Ⅰ、MHC-Ⅱ、Pax7、Pax3表達(dá)的三維分布的研究

【摘要】: 目的通過組織學(xué)、免疫組織化學(xué)及免疫印跡等方法對咽、食管肌層進(jìn)行研究,揭示咽、食管肌層的結(jié)構(gòu)特性及三維分布,為臨床提供理論依據(jù),探討咽、食管肌層發(fā)育和功能性胸痛的潛在機(jī)制。方法①對10例22周胎兒咽、食管及腓腸肌標(biāo)本行HE染色和一抗為α-平滑肌肌動蛋白α-Smooth Muscle Actin,α-SMA㖞、快肌骨骼肌重鏈fast myosin heavy chain,MHC-II㖞、慢肌骨骼肌重鏈slow myosin heavy chain,MHC-Ⅰ㖞、pax7以及pax3抗體的免疫組化SABC染色和免疫印跡。結(jié)果①胎兒咽及食管的組織學(xué)連續(xù)切片能原位、精確地顯示咽段、食管肌纖維的總體構(gòu)型變化,免疫組化染色切片能清楚地區(qū)分咽段、食管連續(xù)肌纖維的類型及其變化規(guī)律。咽部肌纖維表達(dá)Pax7、MHC-I、MHC-II,而不表達(dá)α-SMA。a-SMA、MHC-I、MHC-II共存于胎兒食管上、中段。Pax7、MHC-I、MHC-II共定位,并主要表達(dá)在外部縱行肌,至食管下段逐漸消失。而a-SMA則在食管上端最內(nèi)層開始表達(dá),呈環(huán)行排列,由上至下逐漸增加,并由內(nèi)肌層擴(kuò)展到外肌層。Pax3在咽部和食管均陰性表達(dá)。同時選用腓腸肌作為陽性對照標(biāo)本;結(jié)果Pax7、Pax3共定位于腓腸肌,且Pax7的表達(dá)強(qiáng)于Pax3。②免疫印跡結(jié)果顯示Pax7、MHC-I、MHC-II共定位于咽、食管的肌層,至食管下端逐漸消失。α-SMA則在食管上端開始表達(dá),由上至下逐漸增加。 結(jié)論①胎兒咽、食管骨骼肌的分化沿著由上至食管下端的順序分化。②Pax7、MHC-I、MHC-II共存于胎兒咽、食管上、中段,支持了Pax7陽性細(xì)胞是咽、食管骨骼肌肌祖細(xì)胞的假設(shè)。③進(jìn)一步認(rèn)識了咽、食管骨骼肌和平滑肌的過渡區(qū)。以上結(jié)論將為人類咽、食管的發(fā)育提供新的資料,它將有助于功能性胸痛的臨床診斷,并為球囊擴(kuò)張實驗定位提供進(jìn)一步的理論依據(jù)。
