維甲酸和刺猬因子促進(jìn)骨髓基質(zhì)細(xì)胞向運動神經(jīng)元發(fā)育的實驗研究

【摘要】:本實驗共分為兩部分研究維甲酸(RA)和刺猬因子(Shh)單獨和聯(lián)合誘導(dǎo)對促進(jìn)骨髓基質(zhì)細(xì)胞(BMSCs)向運動神經(jīng)元(MN)發(fā)育的作用。第一部分使用本實驗室制神經(jīng)干細(xì)胞培養(yǎng)基(專利號:ZL02134314.4)培養(yǎng)骨髓基質(zhì)細(xì)胞(BMSCs),掌握BMSCs的原代培養(yǎng)和純化方法,觀察細(xì)胞生長、發(fā)育和分化情況,并通過檢測神經(jīng)干細(xì)胞標(biāo)記Nestin和MN發(fā)育過程重要標(biāo)記蛋白Hb9,研究了BMSCs的增殖、分化生物學(xué)特點。第二部分研究依誘導(dǎo)劑不同分四組(空白組、RA組、Shh組和RA+Shh組)誘導(dǎo)BMSCs,,觀察骨髓源性神經(jīng)干細(xì)胞(BMSCs-D-NSCs)的生長增殖及分化情況,檢測Nestin和Hb9標(biāo)記表達(dá),并結(jié)合發(fā)育神經(jīng)生物學(xué)的知識探討B(tài)MSCs-D-NSCs進(jìn)一步分化為運動神經(jīng)元的潛力。 第一章 小鼠骨髓基質(zhì)細(xì)胞的培養(yǎng)及生物學(xué)特性觀察 目的 研究BMSCs取材、原代培養(yǎng)方法,觀察細(xì)胞在特制培養(yǎng)基的生長、增殖和形態(tài)變化狀況,掌握BMSCs-D-NSCs體外培養(yǎng)要點和生長生物學(xué)特性,獲得穩(wěn)定純化的BMSCs-D-NSCs,為后續(xù)誘導(dǎo)分化實驗奠定基礎(chǔ)。
[Abstract]:This experiment was divided into two parts to study the effects of (RA) and (Shh) on the development of (BMSCs) motoneuron (MN) in bone marrow stromal cells. The first part was to use the culture medium of neural stem cells (patent no.: ZL02134314.4) to culture bone marrow stromal cells (BMSCs),) to master the primary culture and purification methods of BMSCs, and to observe the growth, development and differentiation of the cells. The biological characteristics of proliferation and differentiation of BMSCs were studied by detecting neural stem cell labeled Nestin and Hb9, an important marker in the development of MN. The second part was to observe the growth, proliferation and differentiation of bone marrow derived neural stem cells (BMSCs-D-NSCs) by BMSCs, induced by BMSCs, and to detect the expression of Nestin and Hb9 markers in four groups (control group, RA group, Shh group and RA Shh group). Combined with the knowledge of developmental neurobiology, the potential of BMSCs-D-NSCs to differentiate into motor neurons was discussed. The first chapter: culture and biological characteristics of mouse bone marrow stromal cells The study of BMSCs material, The primary culture method was used to observe the growth, proliferation and morphological changes of the cells in the special medium, to master the main points of BMSCs-D-NSCs culture in vitro and to obtain the stable and purified BMSCs-D-NSCs, which laid the foundation for the subsequent induction and differentiation experiment.
