外周血單個核細(xì)胞在不同條件下HSP70形成的研究

【摘要】:目的:熱休克蛋白(hot shock protein ,HSP)又稱作分子伴侶,是所有細(xì)胞能在應(yīng)激情況下所產(chǎn)生的一組蛋白質(zhì)。根據(jù)分子量的大小分為 HSP20、HSP70、HSP90 和大分子 HSP 家族。HSP70 是 HSP 家族中最重要的一種。HSP70 被認(rèn)為同時與新生的多肽鏈結(jié)合部分,HSP70 作為防止蛋白質(zhì)的聚集及錯誤折疊的媒介。HSP70 與機(jī)體的生理作用密切相關(guān),參與細(xì)胞內(nèi)的許多功能,如蛋白質(zhì)折疊、運(yùn)轉(zhuǎn)、成熟及降解。目前對 HSP 的研究成為熱點(diǎn),但 HSP 的確切作用還仍未了解,特別是介質(zhì)對熱休克蛋白形成的影響研究未見報(bào)道。本試驗(yàn)的目的是通過人外周血單個核細(xì)胞不同時間內(nèi)在不同 pH 和不同介質(zhì)中 HSP70 形成的研究,探討影響熱休克蛋白形成的因素。 方法:抽取無發(fā)燒、無炎癥的健康志愿者外周血,應(yīng)用密度梯度法分離單個核細(xì)胞,分別在不同 pH 的 AB 型血清和不同介質(zhì)(1640 培養(yǎng)液、AB 型人血清、PBS 緩沖液)中 42℃水浴中熱休克 0.5、1 小時,用免疫組化法測定熱休克蛋白 70 的形成。 結(jié)果:外周血單個核細(xì)胞在不同pH的AB型血清中42℃熱休克0.5、1 小時,熱休克蛋白 70 的表達(dá)與未作熱休克處理的空白比較有明顯差異,P0.01。外周血單個核細(xì)胞在 AB 血清中(pH7.35)42℃熱休克0.5、1 小時熱休克蛋白 70 的形成有明顯差異,P0.01。熱休克 0.5小時的表達(dá)明顯高于 1 小時;酸性(pH6.95)AB 血清中 42℃熱休克 0.5、1 小時熱休克蛋白 70 的形成無明顯差異,P0.05;堿性(pH7.65)AB血清中 42℃熱休克 0.5、1 小時熱休克蛋白 70 的形成無明顯差異,P 0.05。 外周血單個核細(xì)胞在不同介質(zhì)中 42℃熱休克 0.5、1 小時,熱休克蛋白 70 的表達(dá)與空白比較有顯著性差異,P0.01。在磷酸鹽緩沖液中42℃熱休克0.5、1小時,熱休克蛋白70的形成有明顯差異,P0.01,0.5 小時的表達(dá)高于 1 小時;1640 培養(yǎng)液中 42℃熱休克 0.5、1 小時,熱休克蛋白 70 的表達(dá)差異不明顯,P0.05。 結(jié)論:(1)外周血單個核細(xì)胞熱休克蛋白 70 形成的量與應(yīng)激源數(shù)量密切相關(guān),多種應(yīng)激源刺激熱休克蛋白質(zhì) 70 形成的量比單一應(yīng)激源刺激形成的快。(2)堿性條件下外周血單個核細(xì)胞也可產(chǎn)生熱休克蛋白。(3)外周血單個核細(xì)胞熱休克蛋白的形成與細(xì)胞所處的介質(zhì)密切相關(guān)。
[Abstract]:Objective: heat shock protein (hot shock protein), also known as molecular chaperone, is a group of proteins produced by all cells under stress. HSP20,HSP70,HSP90 and macromolecule HSP family. HSP70 is the most important type of HSP family. HSP70 is considered to be the medium to prevent protein aggregation and misfolding. Closely related to the physiological function of the body, Participate in many cellular functions, such as protein folding, translocation, maturation, and degradation. At present, the research on HSP has become a hot topic, but the exact role of HSP has not been understood, especially the effect of medium on the formation of heat shock proteins has not been reported. The purpose of this study was to investigate the factors affecting the formation of heat shock proteins (HSPs) in human peripheral blood mononuclear cells (PBMCs) by studying the formation of HSP70 in different pH and medium at different time. Methods: mononuclear cells were isolated from the peripheral blood of healthy volunteers without fever and inflammation by density gradient method.
