SHIV-XJ02170病毒感染中國恒河猴的實驗研究

【摘要】:SHIV病毒艾滋病動物模型是目前國外艾滋病模型研究的趨勢,被認為是比現(xiàn)階段普遍應用的SIV模型針對性更強、更有意義的動物模型。為了盡早開展國內(nèi)此項研究,,尤其是選定毒力較強的SHIV毒株,成功建立SHIV感染動物模型,在前期SIV/SAIDS模型研究的基礎上,本研究以SHIV-XJ02170病毒(以SIVmac239病毒基因組和C型HIV-1病毒的env基因為主體構建的一株SHIV病毒)及前病毒質(zhì)粒接種中國恒河猴,進行了包括臨床表現(xiàn)、病毒學、免疫學等方面研究。為了更準確定量測定實驗恒河猴病毒感染狀態(tài),建立了測定SIV/SHIV病毒RNA載量的SYBR Green Ⅰ熒光染料實時定量RT-PCR方法和測定SIV/SHIV前病毒DNA的SYBR Green Ⅰ實時定量PCR方法。同時為了進一步增強SHIV-XJ02170病毒的毒力,對其進行中國恒河猴體內(nèi)傳代,以期獲得一株高毒力、高致病性的SHIV病毒。 本研究共選用了13只中國恒河猴,實驗前經(jīng)血清學和PCR證明無SIV,STLV-1,SRV/D和B病毒感染。實驗在生物安全三級實驗室中進行。第一代實驗猴共4只,分兩組。第一組2只經(jīng)肌肉注射SHIV-XJ02170前病毒質(zhì)粒DNA;第二組兩只靜脈注射SHIV-XJ02170病毒液。當?shù)谝淮鷮嶒灪餀z出病毒達高峰期后,取10mL感染猴的全血,抗凝后靜脈接種第二代4只實驗猴。第二代實驗猴檢出病毒后再將其全血靜脈接種第三代3只實驗猴,連續(xù)傳至第4代2只猴。每代實驗猴均定期采集肝素和EDTA抗凝血液標本,分別進行病毒分離、PCR方法檢測感染猴PBMC中前病毒DNA、RT-PCR方法檢測血漿病毒RNA、CD4~+/CD8~+、血漿P27抗原等指標的測定,以監(jiān)測SHIV-XJ02170病毒在恒河猴體內(nèi)復制和傳代情況。實驗結果顯示原代SHIV-XJ02170病毒和前病毒質(zhì)粒均可感染中國恒河猴,但傳代的實驗猴病毒檢出率降低,病毒血癥持續(xù)時間明顯縮短,且全部13只實驗猴都未產(chǎn)生典型的艾滋病癥狀,只有二代猴A0408
