姜黃素誘導(dǎo)腫瘤細(xì)胞自噬后的自噬體濃縮疫苗抗腫瘤的機(jī)制研究

【摘要】:目的研究姜黃素誘導(dǎo)腫瘤細(xì)胞自噬后的自噬體濃縮疫苗抗腫瘤的機(jī)制。方法細(xì)胞自噬通過Western blotting和透射電子顯微鏡。小鼠骨髓來源的樹突狀細(xì)胞(BMDCs)的成熟表型經(jīng)常規(guī)掃描電子顯微鏡(SEM)、流式細(xì)胞儀(FCM)和透射電子顯微鏡(TEM)證實(shí)。自噬體介導(dǎo)的交叉呈遞效率通過羥基熒光素二醋酸鹽琥珀酰亞胺脂(CFSE)的分布測定T細(xì)胞的增殖。分析自噬體的抗原交叉呈遞的機(jī)理。結(jié)果姜黃素誘導(dǎo)的自噬通過自噬體的電子顯微鏡以及LC3-Ⅱ的表達(dá)所證實(shí)。自噬體濃縮疫苗上調(diào)表達(dá)CD40、CD86和MHCⅡ。經(jīng)自噬體介導(dǎo)的T細(xì)胞激活后,T細(xì)胞有明顯的擴(kuò)增同時(shí)能殺傷靶細(xì)胞。HSP90、HSP70和HMGB1在這種新的疫苗中發(fā)揮重要作用。結(jié)論這些數(shù)據(jù)表明以自噬體為基礎(chǔ)的疫苗有抗腫瘤療效獨(dú)特的特點(diǎn)。這種新的腫瘤疫苗的功效需臨床試驗(yàn)的進(jìn)一步驗(yàn)證。
