高原創(chuàng)傷性顱腦損傷大鼠液壓模型的實驗研究

【摘要】: 目的:為建立動物顱腦損傷實驗平臺,提高研究質(zhì)量,本研究擬根據(jù)液壓傳遞實驗原理,設(shè)計制作液壓打擊裝置,并明確操作規(guī)范;以此建立可以量化分級的液壓損傷大鼠創(chuàng)傷性顱腦損傷(TBI)模型;對高原環(huán)境下液壓損傷大鼠TBI動物模型進行初步的研究。 方法:1、依據(jù)液壓傳遞原理及加速性顱腦損傷原理,設(shè)計制作能根據(jù)需要調(diào)節(jié)輸出不同打擊力度的動物液壓損傷打擊裝置;2、建立可以分級的液壓損傷大鼠TBI模型:32只成年SD大鼠分為2組:對照組(n=8)和損傷組(n=24)。損傷組又分3個亞組,每亞組各8只大鼠。分別給予0.5-1.0atm、1.5-2.0atm、3.0-3.5atm的壓強打擊致傷后作動物行為和病理學(xué)評估。3、不同海拔環(huán)境下大鼠顱腦液壓損傷后比較觀察:60只成年SD大鼠分為2組:低海拔組(n=30)和高海拔組(n=30)。每組分為對照組(n=6)和損傷組(n=24);每個損傷組按時間點又分4個亞組,每亞組各6只大鼠。損傷組和對照組都進行規(guī)范化的手術(shù)步驟。對照組不進行液壓打擊致傷,損傷組給予約1.5atm打擊壓強造成中度液壓損傷大鼠TBI模型,按時間點分別處理大鼠并取樣,比較不同海拔環(huán)境條件下顱腦損傷動物行為學(xué)及病理學(xué)改變,并觀察其微循環(huán)的變化特征。 結(jié)果:本研究成功制作了動物液壓打擊裝置,在擺錘釋放的角度逐漸增加時,產(chǎn)生的壓強也逐漸增加,而同一角度釋放擺錘產(chǎn)生的壓強基本一致。利用該液壓打擊裝置建立了液壓損傷大鼠TBI動物模型,在不同的打擊壓強下造成的病理改變不同,隨著打擊壓強的增加,造成大鼠的神經(jīng)運動功能障礙,組織病理改變,腦組織含水量的異常也加重。與平原環(huán)境組相比,高原環(huán)境下液壓打擊造成的TBI大鼠病理改變、神經(jīng)功能障礙明顯;腦含水量和vWF改變與平原環(huán)境組相比差異具有統(tǒng)計學(xué)顯著性。 結(jié)論: 1、本研究制作的液壓打擊裝置性能穩(wěn)定,操作簡便,可以較好的應(yīng)用于動物的顱腦損傷實驗研究。2、利用該液壓打擊裝置建立的液壓損傷大鼠TBI模型,可以較好的模擬臨床TBI改變。3、本實驗首次建立了高原環(huán)境下液壓損傷大鼠TBI模型,為進一步開展高原顱腦損傷研究提供了一個可靠的實驗平臺。
[Abstract]:Objective: in order to establish the experimental platform of animal craniocerebral injury and improve the research quality, according to the principle of hydraulic transmission experiment, this study designed and made hydraulic hitting device, and defined the operation standard; The (TBI) model of traumatic craniocerebral injury in rats with hydraulic injury was established, and the TBI animal model of hydraulic injury in high altitude environment was preliminarily studied. Methods: according to hydraulic transfer principle and accelerated craniocerebral injury principle, An animal hydraulic injury attack device was designed and manufactured according to the need to adjust the output of different attack forces. The TBI model of rats with hydraulic injury was established. 32 adult SD rats were divided into two groups: control group (nong8) and injury group (nong24), which could be divided into two groups: control group (n = 8) and injury group (n = 24). The injury group was divided into 3 subgroups with 8 rats in each subgroup. The animal behavior and pathology were evaluated by 0.5-1.0atmAm 1.5-2.0atm-1. 0-3.5atm. After craniocerebral hydraulic injury, 60 adult Sprague-Dawley rats were divided into two groups: low altitude group (nun30) and high altitude group (nun30), and the rats were divided into two groups: low altitude group (n = 30) and high altitude group (n = 30). Each group was divided into two groups: control group (n = 6) and injury group (n = 24), and each group was divided into 4 subgroups (n = 6) and each subgroup (n = 6). Both the injury group and the control group underwent standardized surgical procedures. The TBI model of moderate hydraulic injury was induced by 1.5atm shock pressure in the injury group. The rats were treated and sampled according to the time points. To compare the behavioral and pathological changes of craniocerebral injury at different altitude, and to observe the characteristics of microcirculation. Results: the animal hydraulic impact device was successfully manufactured. When the angle of the pendulum hammer release increased gradually, the pressure produced by the pendulum hammer increased gradually, and the pressure produced by the same angle release pendulum hammer was basically the same. The TBI animal model of rats with hydraulic injury was established by using the device. The pathological changes were different under different impact pressures. With the increase of the impact pressure, the neurological motor dysfunction and histopathological changes were induced in rats. The abnormality of water content in brain tissue is also aggravated. Compared with plain environment group, the pathological changes of TBI rats caused by hydraulic attack under high altitude environment were obvious, and the changes of brain water content and vWF were significantly different from those of plain environment group. Conclusion: 1, the hydraulic hitting device made in this study is stable in performance, easy to operate, and can be used in the experimental study of animal craniocerebral injury. The TBI model of rat with hydraulic injury was established by using the hydraulic hitting device. The model of TBI in rats with hydraulic injury under high altitude environment was established for the first time, which provided a reliable experimental platform for further research on craniocerebral injury at high altitude.
