慢性阻塞性肺疾病寒飲蘊(yùn)肺證大鼠模型的評價(jià)

【摘要】: 目的:從理論上,對慢性阻塞性肺疾病寒飲蘊(yùn)肺證進(jìn)行系統(tǒng)的闡釋與論證,以豐富寒飲蘊(yùn)肺證理論。利用熏煙、氣管滴注脂多糖加寒涼刺激的方法建立慢性阻塞性肺疾病寒飲蘊(yùn)肺證大鼠模型。對大鼠模型可信性作出評價(jià);同時(shí),觀察溫陽化飲方和布地奈德對大鼠慢性阻塞性肺疾病寒飲蘊(yùn)肺證模型干預(yù)的效果。 方法:依據(jù)“外內(nèi)合邪”、“形寒飲冷則傷肺”等觀點(diǎn),結(jié)合慢性阻塞性肺疾病致病原因,在冬季寒冷環(huán)境中冷凍大鼠,大鼠飲水給以冰水,配以熏煙、氣管滴注脂多糖的方法,建立慢性阻塞性肺疾病寒飲蘊(yùn)肺證大鼠模型。觀察其自愈情況,并用溫陽化飲方和布地奈德干預(yù)。觀察大鼠的一般情況、肺組織病理學(xué)改變,檢測有關(guān)肺功能指標(biāo)和血清白介素-8、白介素-10。 結(jié)果:慢性阻塞性肺疾病寒飲蘊(yùn)肺證大鼠模型的一般情況與寒飲蘊(yùn)肺證的臨床表現(xiàn)相符,各項(xiàng)指標(biāo)的變化(除血清白介素-8和白介素-10外)也與慢性阻塞性肺疾病的指標(biāo)變化相一致:大鼠體重減輕,吸氣阻力和呼氣阻力升高、肺順應(yīng)性下降,但血清白介素-8升高和血清白介素-10濃度與正常組相比無顯著性變化(P0.05),病理切片顯示肺組織出現(xiàn)炎性損傷。在撤除造模因素4周后,慢性阻塞性肺疾病寒飲蘊(yùn)肺證模型大鼠的一般情況及各項(xiàng)指標(biāo)沒有明顯好轉(zhuǎn)。用溫陽化飲方和布地奈德干預(yù)的模型大鼠癥狀好轉(zhuǎn),肺功能改善,血清白介素-8和血清白介素-10濃度與慢性阻塞性肺疾病寒飲蘊(yùn)肺證模型大鼠的相比無顯著性變化(P0.05),病理切片示炎性損傷明顯減輕。 結(jié)論:用本實(shí)驗(yàn)方法建立慢性阻塞性肺疾病寒飲蘊(yùn)肺證大鼠模型是可行的。溫陽化飲方和布地奈德在改善慢性阻塞性肺疾病寒飲蘊(yùn)肺證模型大鼠的一般情況、肺功能和抑制氣道炎癥反應(yīng)方面有比較肯定的療效,二者療效沒有差別。
[Abstract]:Objective: in order to enrich the theory of cold drink accumulation lung syndrome in chronic obstructive pulmonary disease (COPD). The model of chronic obstructive pulmonary disease (COPD) was established by fumigation, endotracheal instillation of lipopolysaccharide and cold stimulation. To evaluate the credibility of rat model and observe the effect of Wenyanghua decoction and budesonide on the model of chronic obstructive pulmonary disease (COPD). Methods: according to the viewpoint of "external internal and external evil" and "cold form cold drink hurt lung", combined with the cause of chronic obstructive pulmonary disease (COPD), the rats were given ice water and fumigation in the cold environment in winter. The rat model of chronic obstructive pulmonary disease (COPD) was established by endotracheal instillation of lipopolysaccharide (LPS). Observe its self-healing, and use Wenyang decoction and budesonide intervention. Observe the general situation of rats, lung histopathological changes, detection of lung function indicators and serum interleukin-8, interleukin-10. Results: the general situation of the rat model of cold drink accumulation of lung syndrome in chronic obstructive pulmonary disease was consistent with the clinical manifestation of cold drink accumulation of lung syndrome. The changes (in addition to serum interleukin-8 and interleukin-10) were also consistent with the changes in chronic obstructive pulmonary disease: weight loss, increased inspiratory and expiratory resistance, and decreased lung compliance. However, the increase of serum interleukin-8 and the concentration of serum interleukin-10 had no significant change compared with the normal group (P0.05). Pathological sections showed inflammatory injury in lung tissue. After removing the model factors for 4 weeks, there was no obvious improvement in the general situation and various indexes of the model rats of chronic obstructive pulmonary disease model of cold drink accumulation of lung syndrome. The model rats treated with Wenyanghua decoction and budesonide improved their symptoms and lung function. Serum interleukin-8 and serum interleukin-10 levels were not significantly changed compared with chronic obstructive pulmonary disease model rats (P0.05), pathological sections showed that inflammatory injury was significantly alleviated. Conclusion: it is feasible to establish the model of chronic obstructive pulmonary disease (COPD) with cold drink syndrome. Wenyanghua decoction and budesonide have positive effects on improving the general situation, pulmonary function and inhibiting airway inflammation in rats with chronic obstructive pulmonary disease (COPD) model of cold drink accumulation of lung syndrome, but there is no difference between the two.
