抗甲3型流感病毒卵黃抗體的研究

【摘要】: 流行性感冒是由流感病毒(Influenza virus)引起的急性呼吸道傳染病,由于流感病毒易變異的特性,新型流感病毒的出現(xiàn)常引起該病不同程度的流行。是我國當(dāng)前流行最為廣泛、危害性最嚴(yán)重的一種傳染病。 從本世紀(jì)中葉開始,人們發(fā)現(xiàn)鳥類胚胎及新生兒能從母體獲得大量的IgG抗體,雞蛋蛋黃中也含有大量來源于血清的免疫蛋白IgG,且含量遠(yuǎn)高于血清,被命名為雞卵黃免疫蛋白(Egg yolk immunoglobulin,IgY)。本實(shí)驗(yàn)采用H3N2型流感病毒通過兩種途徑免疫母雞,以水稀釋-硫酸銨沉淀及離子交換層析法提純,并進(jìn)行理化性質(zhì)分析和動(dòng)物效力試驗(yàn)型制備出高效價(jià)的抗甲3型流感病毒IgY.卵黃抗體制備方法簡單,成本低,使用方便,且可廣泛應(yīng)用于人類及禽類疾病的預(yù)防,診斷和治療,具有廣闊的市場(chǎng)開發(fā)和綜合利用的前景。
[Abstract]:Influenza is an acute respiratory infectious disease caused by influenza virus (Influenza virus). Is our country at present the most widespread, the harm most serious kind of infectious disease. Since the middle of this century, it has been found that birds embryos and newborns can obtain a large amount of IgG antibodies from their mothers, and egg yolks also contain a large number of immunoglobulin (IgG) from serum, which is much higher than that of serum. It was named as Egg yolk immunoglobulins (IGY) of chicken egg yolk. In this experiment, hens were immunized with H3N2 influenza virus through two ways, purified by water dilution ammonium sulfate precipitation and ion exchange chromatography, and the high titer anti-influenza virus IgY was prepared by physicochemical properties analysis and animal potency test. The preparation method of yolk antibody is simple, low cost, easy to use, and can be widely used in the prevention, diagnosis and treatment of human and poultry diseases. It has broad market development and comprehensive utilization prospects.
