高糖飼料對大鼠糖脂代謝及肝臟代謝綜合征相關(guān)基因2表達的影響

【摘要】:近年來的研究發(fā)現(xiàn),代謝綜合征(metabolic syndrome,MS)是動脈粥樣硬化(atherosclerosis,AS)的一種新的危險因子。研究表明MS的基本代謝特征是糖、脂代謝異常,但這些糖、脂代謝異常改變發(fā)生的機制以及在MS和AS的發(fā)生發(fā)展中所起作用尚未闡明。本研究室曾采用高糖飼料喂養(yǎng)大鼠建立了一種動物模型,該模型的基本特點是空腹血糖、甘油三酯(total triglyceride,TG)升高;并呈現(xiàn)明顯高凝狀態(tài),明顯具有MS的一些基本特征。將該模型大鼠肝臟mRNA與普通飼料喂養(yǎng)大鼠肝臟mRNA消減雜交,建立了該模型的肝臟差異表達基因cDNA文庫,并對文庫進行篩選,,找到了一些差異表達序列標簽(expressed sequence tag,EST)。將EST與GenBank比對后尋找到與其中一條EST同源的大鼠基因,因為國內(nèi)外尚無該基因結(jié)構(gòu)和功能的報道。我們暫時將該基因命名為代謝綜合征相關(guān)基因2(metabolic syndrome relative gene 2,MSRG2)。本研究在過去工作基礎(chǔ)上進一步探討高糖飼料對大鼠血糖、血脂、肝臟脂質(zhì),特別是血漿及肝臟磷脂及其分布的影響,并測定高糖飼料對大鼠肝臟MSRG2表達的影響。 平衡飼料喂養(yǎng)1周后,實驗組大鼠喂飼高糖飼料,對照組喂飼普通飼
[Abstract]:In recent years, metabolic syndrome (MS) has been found to be a new risk factor for atherosclerotic atherosclerosis (ASA). The results showed that the basic metabolic characteristics of MS were abnormal metabolism of sugar and lipid, but the mechanism of the abnormal metabolism of sugar and lipid and its role in the occurrence and development of MS and as had not been clarified. An animal model was established by feeding rats with high sugar diet in our laboratory. The basic characteristics of the model were fasting blood glucose, total triglyceride-TG), and showed obvious hypercoagulability and had some basic characteristics of MS. The subtractive hybridization of mRNA in the liver of the model rat and the normal feed fed rat liver was used to establish the cDNA library of the liver differential expression gene. The library was screened and some differentially expressed sequence tagESTs were found. After comparing EST with GenBank, a rat gene homologous to one of EST was found, because there are no reports on the structure and function of this gene at home and abroad. We temporarily named the gene 2(metabolic syndrome relative gene 2 MSRG 2. In this study, the effects of high glucose diet on blood glucose, blood lipid, liver lipids, especially plasma and liver phospholipid and their distribution in rats were further studied, and the effects of high glucose diet on the expression of MSRG2 in rat liver were determined. After a week of balanced diet, rats in the experimental group were fed with high-sugar diet, while the control group was fed with common feed.
