【摘要】： DNA微陣列已經(jīng)在功能基因組學(xué)研究中獲得了廣泛的應(yīng)用。在人類疾病研究領(lǐng)域,分離出在疾病狀態(tài)下特異的基因表達(dá)變化具有十分重要的研究意義和實(shí)用價(jià)值。如何從眾多差異表達(dá)基因中分離出那些跟疾病形成與發(fā)展相關(guān)的表達(dá)變化,已經(jīng)成為疾病基因表達(dá)譜數(shù)據(jù)分析的一個(gè)關(guān)鍵問題,目前的差異分析方法還不能完全解決這個(gè)問題。 為了解決這個(gè)問題,提出了系統(tǒng)的基因表達(dá)譜差異分析方法。通過整合基因水平差異分析和功能水平差異分析,從基因、功能兩個(gè)層次上研究基因表達(dá)變化與疾病表型的關(guān)聯(lián)�；蛩讲町惙治霾捎昧诵拚腡檢驗(yàn)算法來分析單個(gè)基因的表達(dá)變化,并用顯著性p值來描述變化程度;功能水平差異分析采用了組合的S檢驗(yàn),通過整合基因的功能編目分類信息,利用單個(gè)基因差異表達(dá)的定量信息,研究一組功能相關(guān)基因的一致性表達(dá)變化。 基于上述的分析策略,開發(fā)了基因表達(dá)譜差異分析軟件MageKey(v1.0)。利用MageKey分析一組人類肌萎縮性脊髓側(cè)索硬化癥疾病的基因表達(dá)譜數(shù)據(jù),測(cè)試結(jié)果顯示了這種分析策略能夠有效地建立疾病表型與基因和功能之間的關(guān)聯(lián)。 通過整合功能注釋信息,從基因水平和功能水平研究基因表達(dá)變化與疾病表型的關(guān)聯(lián),這種分析策略能夠快速地將疾病表型與功能失調(diào)及其表達(dá)變化關(guān)聯(lián)起來,幫助生成一些關(guān)于疾病機(jī)制的生物學(xué)假設(shè)。同時(shí)本文的結(jié)果也說明了整合功能注釋信息研究基因表達(dá)譜數(shù)據(jù)的重要性。
[Abstract]:DNA microarrays have been widely used in functional genomics research. In the field of human disease research, it is of great significance and practical value to separate the specific gene expression changes under the condition of disease. How to separate the expression changes related to the disease formation and development in the multiple differentially expressed genes Chemical analysis has become a key problem in data analysis of disease gene expression profiles. The current difference analysis method can not completely solve this problem.
In order to solve this problem, a systematic method of differential analysis of gene expression profiles is proposed. By integrating gene level difference analysis and functional level difference analysis, the relationship between gene expression changes and disease phenotype is studied from two levels of gene and function. Gene level difference analysis uses a modified T test algorithm to analyze single gene. The changes in expression were described and the degree of change was described with significant P values. The functional level difference analysis adopted a combined S test. By integrating the functional cataloging information of the gene and using quantitative information expressed by a single gene, the consistency of a group of functional genes was studied.
Based on the above analysis strategy, MageKey (v1.0) was developed for gene expression profile difference analysis software. The gene expression profiles of a group of human amyotrophic lateral sclerosis diseases were analyzed by MageKey. The results showed that the analysis strategy could effectively establish the association between the phenotype of the disease and the gene and function.
By integrating functional annotation information, the relationship between gene expression changes and disease phenotype is studied from gene level and functional level. This analysis strategy can quickly associate the disease phenotype with dysfunction and its expression changes and help to generate some biologic hypotheses about the disease mechanism. The results also illustrate the integration of the disease. Functional annotation information studies the importance of gene expression data.
