糠秕馬拉色菌胞外蛋白酶活性測定

【摘要】:目的:①檢測糠秕馬拉色菌胞外蛋白酶活性;②研究菌絲相及酵母相蛋白酶活性的差異;③蛋白酶活性的影響因素。 方法:選用糠秕馬拉色菌標準株,接種于菜籽油液體培養(yǎng)基和菌絲液體培養(yǎng)基中振蕩培養(yǎng),,誘導糠秕馬拉色菌菌絲相及酵母相生成,收集培養(yǎng)上清液。用全脂牛奶平板法、Folin-酚法、QuantiCleave蛋白酶試劑盒測定菌絲相及酵母相糠秕馬拉色菌胞外蛋白酶的活力,并檢測溫度、pH值對蛋白酶活性的影響。 結果:①全脂牛奶平板法:培養(yǎng)第7天菌絲相上清液蛋白酶活性(12.5±1.2mm)高于酵母相(9.25±0.71mm)(p<0.05);菌絲相上清液蛋白酶活性隨培養(yǎng)天數(shù)延長而增大;25℃和37℃兩種溫度對上清液蛋白酶活性無明顯影響(P>0.05);②Folin-酚法:上清液蛋白酶活性隨培養(yǎng)天數(shù)、菌落計數(shù)、菌絲誘導率增加而增大,培養(yǎng)液pH值呈下降趨勢;同一天的菌絲相上清液蛋白酶活性明顯高于酵母相(p<0.05);三種溫度(25℃、37℃、49℃)及三種pH值(3.8、5.6、7.4)對上清液蛋白酶活性無明顯影響;溫度37℃、pH5.6時菌絲相上清液蛋白酶活性最高;③QuantiCleave蛋白酶試劑盒:菌絲相及酵母相上清液中均含有蛋白酶且
[Abstract]:Objective to detect the extracellular protease activity of Malassezia furfur by 1: 1 and to study the factors influencing the protease activity in mycelia and yeast. Methods: the standard strain of Malassezia furfuris was inoculated in the liquid medium of rapeseed oil and the liquid medium of hyphae to induce the formation of hyphae and yeast phase of Malassezia furfurum and the supernatant of culture was collected. The extracellular protease activity of Malassezia furfurum in hyphal phase and yeast phase was determined by Folin- phenol QuantiCleave protease kit, and the effect of pH value on the activity of protease was detected.
