【摘要】： 本研究首先從臨床分離陰道毛滴蟲,用肝浸湯進(jìn)行純培養(yǎng),提取蟲體總核酸,1%瓊脂糖凝膠電泳,初步篩選出一株攜帶陰道毛滴蟲病毒的蟲株;其次通過(guò)電鏡觀察和生化分子生物學(xué)方法對(duì)病毒進(jìn)一步進(jìn)行鑒定,首次證實(shí)了在我國(guó)陰道毛滴蟲體內(nèi)存在病毒,核酸大小約為5.5kb,病毒為外觀球形、直徑33nm的dsRNA病毒,α-~(32)P標(biāo)記的UTP測(cè)得該病毒具有RNA依賴RNA聚合酶活性,病毒序列與陰道毛滴蟲病毒T1株序列具有82.9%的同源性;成功構(gòu)建病毒衣殼蛋白基因的原核表達(dá)載體pET-cap2037,并在大腸桿菌BL(DE3)中進(jìn)行了表達(dá)。本研究的結(jié)果為深入研究我國(guó)陰道毛滴蟲病毒奠定了良好基礎(chǔ)。
[Abstract]:In this study, we first isolated Trichomonas vaginalis from clinical practice, used liver dipping Soup for pure culture, extracted insect total nucleic acid, 1% agarose gel electrophoresis, preliminarily screened a strain of Trichomonas vaginalis virus. Secondly, the virus was identified by electron microscope observation and biochemical molecular biology method, and the first confirmation of vaginal dripping in our country was made. The virus has a virus in the body, the size of nucleic acid is about 5.5kb, the virus is a spherical dsRNA virus with a diameter of 33nm, and the UTP of alpha ~ (32) P marks that the virus has RNA dependent RNA polymerase activity. The sequence of the virus is homologous to the sequence of the T1 strain of Trichomonas vaginalis virus, and the prokaryotic expression vector of viral capsid protein gene is constructed. 037, and expressed in E. coli BL (DE3). The results of this study laid a good foundation for further study of Trichomonas vaginalis virus in China.
