【摘要】:雌激素主要通過(guò)與雌激素受體結(jié)合發(fā)揮生物學(xué)作用。在雄性動(dòng)物的生殖系統(tǒng)中有大量的雌激素產(chǎn)生,適當(dāng)濃度的雌激素對(duì)雄性生殖的生長(zhǎng)發(fā)育和維持正常功能有非常重要的作用,雌激素缺乏或過(guò)高濃度的暴露都會(huì)引起對(duì)雄性動(dòng)物生殖系統(tǒng)生長(zhǎng)發(fā)育和功能的損害,但目前對(duì)雌激素在生殖系統(tǒng)中的作用機(jī)制仍然了解不多。由于環(huán)境中的雌激素和有雌激素活性的化學(xué)物質(zhì)不斷增多,使人們迫切的想知道雌激素對(duì)雄性動(dòng)物生殖系統(tǒng)的作用位點(diǎn)和作用機(jī)制以及雄性動(dòng)物和人暴露于生活中的環(huán)境雌激素或雌激素樣物質(zhì)是否會(huì)導(dǎo)致其生殖結(jié)構(gòu)和正常功能受到不利的影響。本實(shí)驗(yàn)?zāi)康氖菫榱颂剿髟谛律诒┞队诖萍に?雄性大鼠生殖系統(tǒng)生長(zhǎng)發(fā)育和功能受到的不利影響及其作用機(jī)制。方法:在出生第2天至12天期間,新生期雄性大鼠分組分別隔日給不同劑量:10μg/2d、500ng/2d和25ng/2d炔雌醇處理(處理組),對(duì)照組大鼠給予玉米油。飼養(yǎng)至成年后,處死,取材檢測(cè)各組成年大鼠睪丸和附睪的一般形態(tài)結(jié)構(gòu)、精子發(fā)生效率和附睪精子數(shù)量的改變,并用RT—PCR法檢測(cè)睪丸內(nèi)精子頂體蛋白酶的表達(dá)變化和用酶組織化學(xué)方法檢測(cè)輸精管內(nèi)精子頂體蛋白酶的活性改變。結(jié)果:炔雌醇處理的10μg組和25ng組的部分大鼠出現(xiàn)隱睪、睪丸積液和附睪腫物;雖然各處理組大鼠成年后的體重與對(duì)照組的相比沒(méi)有差異,但處理組的雙側(cè)睪丸的絕對(duì)重量、相對(duì)重量和雙側(cè)附睪的絕對(duì)重量顯著下降(P0.01)。第Ⅶ、Ⅷ期生精小管管徑大小、小管上皮高度和管腔直徑大小除了25ng組與對(duì)照組沒(méi)有明顯差異外,其余兩組與對(duì)照組和25ng組相比,均有明顯降低(P0.05)。各處理組大鼠的每日精子產(chǎn)生量和附睪精子總數(shù)與對(duì)照組相比沒(méi)有明顯差異(P0.05);但處理組的附睪精子數(shù)量均比對(duì)照組低。給予炔雌醇的劑量越大,頂體蛋白酶呈活性的精子比例就越低,在10μg組、500ng組、25ng組和對(duì)照組中該酶呈活性的精子比例分別為54.72±5.87%、64.93±4.06%、73.78±1.87%和81.64±1.55%,各處理組與對(duì)照組相比有顯著性差異(P0.01)。相對(duì)于對(duì)照組,10μg組和25ng組大鼠睪丸內(nèi)頂體蛋白酶的mRNA表達(dá)也明顯減少(P0.05)。結(jié)論:1)新生期雄性大鼠暴露于炔
[Abstract]:Estrogen plays a biological role mainly by binding with estrogen receptor. A large amount of estrogen is produced in the reproductive system of male animals, and the proper concentration of estrogen plays a very important role in the growth and development of male reproduction and the maintenance of normal function. Estrogen deficiency or exposure to high concentrations of estrogen can cause damage to the growth, development and function of reproductive system in male animals, but the mechanism of estrogen in reproductive system is still not well understood. Because of the increasing amount of estrogen and estrogen-active chemicals in the environment, It is urgent to know the action site and mechanism of estrogen on the reproductive system of male animals and whether the environmental estrogens or estrogen-like substances exposed to life of male animals and people will lead to their reproductive structure and positive. Normal functions are adversely affected. The aim of this study was to explore the adverse effects of estrogen exposure on the growth, development and function of the reproductive system in male rats and its mechanism. Methods: from day 2 to day 12, newborn male rats were treated with different doses of 1: 10 渭 g / 2, 500 ng / 2 d and 25ng/2d ethylestradiol (treatment group, control group) with corn oil. After feeding to adulthood, the rats were sacrificed. The morphological structure of testis and epididymis, the spermatogenesis efficiency and the number of epididymal spermatozoa were measured. The expression of acrosin in spermatozoa and the activity of acrosin in spermatozoa were detected by RT-PCR method and enzyme histochemical method respectively. Results: there were cryptorchidism, testicular effusion and epididymal mass in 10 渭 g group treated with ethylestradiol and part of rats in 25ng group, although the weight of adult rats in each treatment group was not different from that of control group, the absolute weight of bilateral testis in treatment group was not different from that in control group.
