【摘要】：目的:通過快速大容量尾靜脈注射方法構(gòu)建循環(huán)肝癌細(xì)胞肝內(nèi)復(fù)發(fā)轉(zhuǎn)移小鼠模型。方法:40只C57BL/6J小鼠采用隨機(jī)數(shù)字表法隨機(jī)分為對照組和實(shí)驗(yàn)組(20只/組),對照組采用傳統(tǒng)的緩慢小容量尾靜脈注射法(2×106個(gè)肝癌Hepa1-6細(xì)胞/0.2 ml,30 s內(nèi)注射完畢),實(shí)驗(yàn)組采用改良的快速大容量尾靜脈注射法(2×106個(gè)肝癌Hepa1-6細(xì)胞/2 ml,5 s內(nèi)注射完畢)構(gòu)建循環(huán)肝癌細(xì)胞肝內(nèi)復(fù)發(fā)轉(zhuǎn)移小鼠模型。4周后摘取肝、肺、腎、脾組織并行H-E染色,大體和鏡下觀察肝臟、肺臟、脾臟和腎臟的成瘤情況。結(jié)果:對照組小鼠只在肺臟有轉(zhuǎn)移灶形成,成瘤率為95%(19/20),肝臟、腎臟、脾臟未見有轉(zhuǎn)移灶形成。實(shí)驗(yàn)組小鼠在肝臟和肺臟同時(shí)形成轉(zhuǎn)移灶,肺臟成瘤率為94.7%(18/19),肝臟成瘤率為100%(19/19),脾臟、腎臟未見轉(zhuǎn)移灶形成。結(jié)論:快速大容量尾靜脈注射法注射循環(huán)肝癌細(xì)胞構(gòu)建的小鼠模型可以模擬肝癌根治性切除后殘留的循環(huán)肝癌細(xì)胞導(dǎo)致早期肝癌肝內(nèi)復(fù)發(fā)轉(zhuǎn)移的過程。
[Abstract]:Objective: to establish a mouse model of intrahepatic recurrence and metastasis of circulating liver cancer cells by rapid large volume caudal vein injection. Methods 40 C57BL/6J mice were randomly divided into two groups: control group (n = 20) and experimental group (n = 20). The control group (n = 20) was injected with 2 脳 10 6 Hepa1-6 cells per 0.2 ml in 30 s. The mice model of hepatic recurrence and metastasis of circulating hepatoma cells was established by a modified rapid large volume caudal vein injection method of 2 脳 106 hepatoma Hepa1-6 cells / 2 ml / min within 5 seconds. Lung, kidney and spleen were stained with H-E staining. Liver, lung, spleen and kidney were observed under microscope. Results: in the control group, metastasis was only found in the lung, and the tumorigenesis rate was 95% 20%. No metastasis was found in the liver, kidney and spleen. In the experimental group, the metastatic foci were formed in the liver and lung simultaneously, the tumorigenesis rate of the lung was 94.770 / 19, the tumorigenesis rate of the liver was 100 / 19 / 19, the spleen and the kidney were not metastasized. Conclusion: the mouse model established by rapid large volume caudal injection of circulating hepatoma cells can simulate the process of recurrence and metastasis of early liver cancer caused by residual circulating hepatoma cells after radical resection of liver cancer.
