線蟲(chóng)磷脂酶D的生物學(xué)功能研究

【摘要】:磷脂酶D(Phospholipase, PLD)是細(xì)胞信號(hào)轉(zhuǎn)導(dǎo)網(wǎng)絡(luò)中的重要成員之一,與許多人類(lèi)疾病都有密切的關(guān)系,是一個(gè)潛在的藥物治療靶標(biāo)。但到目前為止,其生物學(xué)功能和作用機(jī)制尚未研究清楚,由于人類(lèi)的細(xì)胞信號(hào)轉(zhuǎn)導(dǎo)系統(tǒng)非常復(fù)雜,所以本研究選擇了相對(duì)簡(jiǎn)單的模式生物——線蟲(chóng),來(lái)研究PLD的生物學(xué)功能,并對(duì)其作用機(jī)制進(jìn)行初步的探討。線蟲(chóng)作為一種真核多細(xì)胞生物,其基因組序列的測(cè)定早在1998年就已經(jīng)完成,經(jīng)過(guò)多年研究發(fā)現(xiàn),線蟲(chóng)存在著許多與高等生物類(lèi)似的細(xì)胞信號(hào)轉(zhuǎn)導(dǎo)通路,并且與其它多種生物模型相比,線蟲(chóng)還具有結(jié)構(gòu)簡(jiǎn)單、飼養(yǎng)方便、生命周期短以及便于觀測(cè)等許多優(yōu)點(diǎn)。 在本研究中,我們以PLD-1基因敲除的線蟲(chóng)種系RB1040作為研究對(duì)象,通過(guò)比較其與野生型線蟲(chóng)N2在相同培養(yǎng)條件下表型(產(chǎn)卵數(shù)量、壽命、運(yùn)動(dòng)、吞咽、排便、化學(xué)趨勢(shì)和食物趨勢(shì)等)上的差異來(lái)推測(cè)PLD的生物學(xué)功能。同時(shí)還利用連接有綠色熒光蛋白(GFP)的PLD-1敲除型線蟲(chóng)來(lái)研究PLD-1的表達(dá)部位,初步探討了PLD-1在線蟲(chóng)中的作用機(jī)制,為研究人PLD的生物學(xué)功能提供了理論依據(jù)。
[Abstract]:Phospholipase phospholipase (PLD) is one of the important members of cell signal transduction network and has close relationship with many human diseases and is a potential target for drug therapy.But up to now, its biological function and mechanism have not been studied. Because of the complexity of human cell signal transduction system, we chose a relatively simple model organism, nematode, to study the biological function of PLD.The mechanism of its action was discussed.As a kind of eukaryotic multicellular organism, the genome sequence of nematode has been completed since 1998. After many years of research, it has been found that nematodes have many cell signal transduction pathways similar to those of higher organisms.Compared with other biological models, nematode has many advantages, such as simple structure, convenient feeding, short life cycle and easy observation.In this study, we compared the phenotypes (egg number, longevity, movement, swallowing, defecation) of PLD-1 knockout nematode strain RB1040 under the same culture conditions as wild-type nematode N2.The differences in chemical and food trends are used to speculate on the biological function of PLD.At the same time, the PLD-1 knockout nematodes with green fluorescent protein (GFP) were used to study the expression of PLD-1, and the mechanism of PLD-1 online was discussed, which provided a theoretical basis for the study of the biological function of human PLD.
