【摘要】: 目的: 心肌梗死患者的干細(xì)胞移植治療能夠取代壞死的心肌細(xì)胞、增加有功能的心肌細(xì)胞數(shù)量,從而達(dá)到改善心功能的目的,是一種理想的治療手段。目前,干細(xì)胞治療還處于基礎(chǔ)實(shí)驗(yàn)研究和初期的臨床研究階段,是全球心血管疾病研究者關(guān)注的焦點(diǎn)。P19胚胎癌細(xì)胞是目前研究較成熟的一種胚胎源性干細(xì)胞,深受廣大干細(xì)胞研究者青睞。催產(chǎn)素(Oxytocin, OT)長期以來被認(rèn)為只是一種女性生殖激素,然而,近年來隨著其多方面作用的發(fā)現(xiàn),逐漸被越來越多的研究人員所重視。近年的研究發(fā)現(xiàn),OT還具有促進(jìn)心臟發(fā)生的作用[1]。本實(shí)驗(yàn)應(yīng)用OT作用于培養(yǎng)的P19胚胎源性干細(xì)胞,誘導(dǎo)其向心肌細(xì)胞方向分化,并通過免疫細(xì)胞化學(xué)技術(shù)、激光掃描共聚焦顯微鏡技術(shù)、透射電子顯微鏡技術(shù)等方法對(duì)誘導(dǎo)分化的細(xì)胞進(jìn)行形態(tài)學(xué)觀察,以期為心肌損傷干細(xì)胞治療提供基礎(chǔ)實(shí)驗(yàn)數(shù)據(jù)。 方法: 1 P19細(xì)胞誘導(dǎo)分化 取對(duì)數(shù)生長期的P19細(xì)胞,經(jīng)0.25%胰酶-0.2%EDTA消化,制成單細(xì)胞懸液。調(diào)整細(xì)胞密度為2×10~5/ml,以每皿2ml接種于0.5%瓊脂鋪底的直徑35mm細(xì)菌級(jí)培養(yǎng)皿中懸浮培養(yǎng)。實(shí)驗(yàn)組1、2、3的培養(yǎng)基中分別含OT 1×10~(-7) mol/ L、3×10~(-7) mol/ L、5×10~(-7) mol/ L;對(duì)照組的培養(yǎng)基中不含OT。
[Abstract]:Objective:Stem cell transplantation in patients with myocardial infarction can replace the necrotic cardiomyocytes and increase the number of functional cardiomyocytes so as to improve the cardiac function and is an ideal treatment method.At present, stem cell therapy is still in the stage of basic experimental research and early clinical research, which is the focus of global cardiovascular disease researchers. P19 embryonic cancer cell is a mature embryonic stem cell.It is favored by stem cell researchers.Oxytocin (OT) has long been considered as a female reproductive hormone. However, with the discovery of its various roles, more and more researchers pay more and more attention to it.In recent years, it has been found that OT can also promote the development of the heart [1].In this study, OT was used to induce the differentiation of P19 embryonic stem cells into cardiomyocytes, and by immunocytochemistry, laser scanning confocal microscopy (LSCM) was used to induce the differentiation of P19 embryonic stem cells into cardiomyocytes.Transmission electron microscopy (TEM) was used to observe the morphology of differentiated cells in order to provide experimental data for the treatment of myocardial injury stem cells.Methods:1 P19 cell differentiationP19 cells in logarithmic growth stage were digested by 0.25% trypsin-0.2TA and single cell suspension was prepared.The cell density was adjusted to 2 脳 10 ~ (5) / ml, and the suspension culture was carried out in 0.5% Agar (0.5% Agar) -coated diameter 35mm bacterial-grade culture dish with each dish of 2ml.OT1 脳 10 ~ (10) mol/ L ~ (7) mol/ L ~ (3 脳 10 ~ (-7)) mol/ L ~ (5 脳 10 ~ (-7)) mol/ L were found in the culture medium of the experimental group (1 脳 10 ~ (2) C), but not in the medium of the control group.
