心房顆粒釋放形式和轉(zhuǎn)運(yùn)途徑的結(jié)構(gòu)基礎(chǔ)研究

【摘要】:目的:放射免疫分析表明,心包腔的心包液中,存在心房尿鈉多肽簡(jiǎn)稱(chēng)心房肽(ANP);而且,在生理或病理的情況下,心包液中心房肽的濃度,約為血漿中的4~100倍。這種濃度差提示,心包液中的心房肽,可能由心肌細(xì)胞直接分泌入心包腔。然而,關(guān)于心包腔中存在高濃度心房肽的生理和病理意義,日前仍不十分清楚。對(duì)于心包液中心房肽的來(lái)源和去向問(wèn)題,尚缺乏形態(tài)學(xué)方面的證據(jù)。本研究通過(guò)對(duì)心房和心包的光鏡和電鏡觀察,探討心房顆粒(ASG)或心房肽的釋放形式,研究心房顆;蛐姆侩脑诩(xì)胞外的轉(zhuǎn)運(yùn)途徑,為心包液中心房肽的來(lái)源、去向及其重要意義,提供結(jié)構(gòu)基礎(chǔ)和形態(tài)學(xué)方面的依據(jù)。 方法:(1) 對(duì)家兔右心耳及其對(duì)應(yīng)的心包膜,進(jìn)行蘇木精—伊紅(HE)染色和5’核苷酸酶(5’-Nase)組織化學(xué)染色,應(yīng)用光鏡(LM),觀察心外膜和心包膜的細(xì)微結(jié)構(gòu),以及其中的淋巴管分布特點(diǎn)。 (2) 應(yīng)用透射電鏡(TEM),觀察右心耳和心包膜的超微結(jié)構(gòu),以及上皮細(xì)胞之間連接的特點(diǎn),辨別心肌細(xì)胞之間毛細(xì)血管的類(lèi)型;觀察毛細(xì)淋巴管和心房顆粒的超微結(jié)構(gòu)及其分布特征。 (3) 應(yīng)用掃描電鏡(SEM),觀察心外膜表面和心包膜腔面的結(jié)構(gòu)特點(diǎn);對(duì)NaOH消蝕處理后的心包膜樣品,觀察間皮細(xì)胞下結(jié)締組織纖維的構(gòu)筑特征。
[Abstract]:Objective: radioimmunoassay showed that atrial natriuretic peptide (ANPP) was present in pericardial fluid of pericardium, and that the concentration of atrial peptide in pericardial fluid was 100 times higher than that in plasma under physiological or pathological conditions.This concentration difference suggests that atrial peptides in pericardial fluid may be secreted directly by cardiomyocytes into the pericardial cavity.However, the physiological and pathological significance of the presence of high concentrations of atrial peptides in the pericardium remains unclear.There is no morphological evidence for the origin and whereabouts of atrial peptides in pericardial fluid.The aim of this study was to investigate the release form of atrial granule ASG or atrial peptide by means of light and electron microscope observation of atrium and pericardium, and to study the pathway of translocation of atrial granule or atrial peptide in pericardial fluid, which was the source of atrial peptide in pericardial fluid.The destination and its significance provide the basis of structure and morphology.Methods the right atrial appendage and its corresponding pericardial membrane were stained with hematoxylin eosin (HEH) and 5 'nucleotidase 5' -Nase (Nase) histochemical staining. The fine structure of epicardium and pericardium was observed by light microscope.And the characteristics of lymphatic distribution.The ultrastructure of right atrial appendage and pericardium and the characteristics of the connection between epithelial cells were observed to distinguish the types of capillaries between cardiac myocytes, and the ultrastructure and distribution of capillary lymphatics and atrial granules were observed by means of transmission electron microscope (TEM).The structural characteristics of epicardial surface and pericardial cavity surface were observed by scanning electron microscope (SEM), and the architectural characteristics of connective tissue fibers under mesothelial cells were observed for pericardial samples treated with NaOH.
