【摘要】:【目的】 鑒定淡色庫蚊溴氰菊酯抗性和敏感品系中肌球蛋白輕鏈2基因(NYD-MLC2)的表達差異,獲取NYD-MCL2 cDNA全長序列,,并構建NYD-MLC2原核和昆蟲表達載體,為進一步研究奠定基礎。 【方法】 用熒光定量PCR法鑒定NYD-MLC2在淡色庫蚊溴氰菊酯抗性和敏感品系中的表達差異。用RACE法從抗性品系中擴增NYD-MLC2 cDNA全長序列,對NYD-MLC2編碼的蛋白進行生物信息學分析。用Blast軟件在NCBI和SwissProt數(shù)據庫中進行同源性比對,用Clustalw軟件進行聚類分析,用EMBL pI/Mw tool計算等電點和分子量,Prot-Param分析NYD-MLC2基因編碼的蛋白質的基本性質,Signal3.0軟件進行信號肽分析,ProtScale軟件進行疏水性分析,PSORT Ⅱ軟件進行細胞定位分析,PatSearch軟件進行基序分析,SMART軟件進行蛋白質結構域分析,HNN軟件分析二級結構,Tmpred軟件進行跨膜區(qū)預測等。通過基因重組方法構建NYD-MLC2原核和昆蟲表達載體。 【結果】 熒光定量PCR結果顯示,NYD-MLC2在淡色庫蚊溴氰菊酯抗性品系中的表達量是敏感品系的4.08倍。獲得淡色庫蚊NYD-MLC2 cDNA全長序列,其中完整開讀框序列為630bp,經推導編碼210個氨基酸(GenBank/NCBIDQ140391,2005),與多個物種的肌球蛋白輕鏈2基因比對,同源性大于25%。成功構建NYD-MLC2原核和昆蟲表達載體。 【結論】 NYD-MLC2在淡色庫蚊溴氰菊酯抗性品系中高表達,提示NYD-MLC2與淡色庫蚊溴氰菊酯抗性相關,值得進一步研究。
[Abstract]:[objective] to identify the difference of expression of myosin light chain 2 gene (NYD-MLC2) in Culex pipiens pallens deltamethrin resistance and sensitive strain, obtain the full-length NYD-MCL2 cDNA sequence, and construct NYD-MLC2 prokaryotic expression vector and insect expression vector, and lay a foundation for further study.[methods] the expression of NYD-MLC2 in deltamethrin resistant and sensitive strains of Culex pipiens pallens was identified by fluorescence quantitative PCR.The full-length sequence of NYD-MLC2 cDNA was amplified by RACE from resistant strains, and the protein encoded by NYD-MLC2 was analyzed by bioinformatics.Comparing the homology between NCBI and SwissProt database with Blast software, cluster analysis with Clustalw software and EMBL with EMBLPI/Mw tool calculation of isoelectric points and Molecular weight Prot-Param Analysis of the basic Properties of proteins encoded by NYD-MLC2 Gene; signal peptide analysis by signal peptide 3.0 software; hydrophobicity analysis by ProtScale software; cell localization analysis by PSORT 鈪
