【摘要】:內(nèi)嗎啡肽(Endomorphin,EM)是與μ阿片受體的親和力和選擇性最高的內(nèi)源性配體,可分為EM1和EM2兩種亞型,具有明確的鎮(zhèn)痛作用。EM陽性神經(jīng)元的胞體在中樞內(nèi)主要分布于下丘腦、孤束核等,在外周主要分布于感覺神經(jīng)節(jié),而EM陽性纖維及其終末的分布較為廣泛,但主要存在于下丘腦、孤束核、脊髓和延髓背角等許多重要結(jié)構(gòu)。以往的研究證明EM1陽性神經(jīng)元、纖維及終末主要存在于上位中樞,EM2陽性神經(jīng)元、纖維及終末則主要位于脊髓等下位中樞和感覺神經(jīng)節(jié),提示EM1可能主要在高位中樞發(fā)揮鎮(zhèn)痛作用,而EM2主要參與對傷害性刺激初級傳入的調(diào)控。下丘腦、孤束核是內(nèi)臟感覺信息傳遞的重要通路,并參與痛信息的調(diào)控,這些核團(tuán)之間是否有EM陽性纖維往返聯(lián)系以及它們的EM陽性纖維及終末的來源如何目前仍未見報道。本研究利用熒光金(FG)逆行追蹤與EM免疫熒光組織化學(xué)染色相結(jié)合的雙標(biāo)技術(shù),,觀察了孤束核和下丘腦之間EM能纖維的往返聯(lián)系。將FG注入孤束核(以外側(cè)亞核為中心)后,在下丘腦室旁核小細(xì)胞部、穹窿周區(qū)、下丘腦后區(qū)及弓狀核等核團(tuán)內(nèi)都觀察到數(shù)量較多的FG逆行標(biāo)記神經(jīng)元;EM1和EM2陽性神經(jīng)元的胞體主要位于下丘腦背內(nèi)側(cè)核、腹內(nèi)側(cè)核
[Abstract]:Endomorphine is an endogenous ligand with the highest affinity and selectivity to 渭 opioid receptors. It can be divided into two subtypes: EM1 and EM2. The cell bodies of EM positive neurons are mainly distributed in the hypothalamus, nucleus solitarius and so on.EM-positive fibers and their terminals are widely distributed in the sensory ganglia, but mainly in the hypothalamus, nucleus tractus solitarius, spinal cord and dorsal horn of medulla oblongata and so on.Previous studies have shown that EM1 positive neurons, fibers and terminals are mainly located in the epigastric center of EM2 positive neurons, and the fibers and terminals are mainly located in the spinal cord and the sensory ganglion.The results suggest that EM1 may play an analgesic role in the high central nervous system, while EM2 may be involved in the regulation of nociceptive stimulation of primary afferent.Hypothalamus and nucleus solitarii are important pathways of visceral sensory information transmission, and participate in the regulation of pain information. Whether there is an EM-positive fiber connection between these nuclei, and how their EM positive fibers and their source of terminal are still unknown.In this study, the relationship between EM energy fibers in nucleus solitarius and hypothalamus was observed by using the double labeling technique of fluorescence FG retrograde tracing and EM immunofluorescence histochemical staining.After FG was injected into the nucleus of the solitary tract (with the lateral subnucleus as the center), the paraventricular nucleus of the hypothalamus, the perifornical area, was located in the paraventricular nucleus of the hypothalamus.In the posterior hypothalamus and arcuate nucleus, a large number of FG retrograde labeled neurons EM1 and EM2 positive neurons were observed in the dorsal medial hypothalamus and ventromedial nucleus.
