綜合放血法致小鼠血虛證模型的相關(guān)研究

【摘要】:血虛證是中醫(yī)臨床常見證侯,但是建立貼近于中醫(yī)臨床的藥理血虛證模型的文獻(xiàn)報(bào)道很少。本課題是在采用 (60)~Co γ射線照射和環(huán)磷酰胺腹腔注射制作小鼠血虛證模型基礎(chǔ)之上,通過慢性放血,限食,游泳建立了綜合放血法血虛證模型,并分別從外周血象、細(xì)胞周期、(CD34)~+ 細(xì)胞數(shù)量、造血祖細(xì)胞的集落生成能力、骨髓和脾臟細(xì)胞凋亡等方面,探討了綜合放血法小鼠血虛證發(fā)生發(fā)展的機(jī)制,旨在完善不同方法建立血虛證模型的實(shí)驗(yàn)數(shù)據(jù),從而全面,客觀的評(píng)價(jià)中醫(yī)血虛證模型的病變規(guī)律。四物湯是一補(bǔ)血活血調(diào)經(jīng)的經(jīng)典名方,由熟地黃、當(dāng)歸、白芍、川芎組成,具有確切的療效。本研究較系統(tǒng)地觀察了四物湯對(duì)血虛證模型小鼠的防治作用,闡明了四物湯對(duì)綜合放血法血虛證模型的補(bǔ)血作用的分子機(jī)制。 本研究通過每日按 75g.kg~(-1) 體重限食,隔日游泳 20min 及放血 0.4ml 建立綜合放血法小鼠血虛證模型,造模共 15d。實(shí)驗(yàn)結(jié)果顯示:小鼠在造模第 9d 左右出現(xiàn)皮毛干燥,光澤差,懶動(dòng)等不同程度的血虛癥象;外周血象紅細(xì)胞總數(shù)及血紅蛋白自造模之日起至第 14d 造模結(jié)束都是極顯著降低;其 (CD34)~+ 細(xì)胞比例在第 7,12d 顯著高于正常組;骨髓細(xì)胞 G0/1期細(xì)胞數(shù)在第 4,7,21d 極顯著低于正常組;而其 S 期細(xì)胞數(shù)和增殖指數(shù)在第 4,7,21d 極顯著高于正常組;胸腺及胸腺指數(shù)顯著降低。 為探討四物湯預(yù)防和治療綜合放血法小鼠血虛證的作用機(jī)理,分別在制作模型前后給小鼠灌胃給藥。結(jié)果表明:四物湯治療給藥促進(jìn)骨髓細(xì)胞的增殖;抑制骨髓造血干祖細(xì)胞的凋亡;增強(qiáng)血虛小鼠的免疫功能。四物湯預(yù)防給藥的作用更加明顯,對(duì)小鼠骨髓造血干祖細(xì)胞、外周血細(xì)胞的恢復(fù)有明顯促進(jìn)作用;有減少骨髓細(xì)胞G2/M阻滯,保護(hù)骨髓和促進(jìn)骨髓細(xì)胞由 G1期進(jìn)入 S 期的效應(yīng)。促進(jìn)骨髓造血干祖細(xì)胞的增殖能力,抑制骨髓造血干祖細(xì)胞的凋亡可能是四物湯治療綜合放血法血虛證的機(jī)制之一。 綜合放血法,(60)~Coγ 射線照射,環(huán)磷酰胺腹腔注射建立的血虛證模型的比較實(shí)驗(yàn)結(jié)果顯示:綜合放血法組紅細(xì)胞和血紅蛋白降低明顯,(60)~Co γ射線照射組白細(xì)胞降低明顯,環(huán)磷酰胺組白細(xì)胞和紅細(xì)胞降低明顯;綜合放血組在造模第 15d,(60)~Co γ射線照射組在造模后 7d,環(huán)磷酰胺組在造模后第 7d 的 CFU-E,BFU-E,CFU-GM 均顯著降低;綜合放血組 (CD34)~+ 細(xì)胞在第 8,11d 顯著升高,(60)~Coγ 射線照射組 CD34+細(xì)胞自造模后至第 14d 均顯著低于正常組,環(huán)磷酰胺組從造模后先升高后降低;三種模型的胸腺指數(shù)均顯著低于正常組,但是綜合放血組脾臟指數(shù)顯著高于正常組而射線照射組相反。 總的來說,綜合放血法指標(biāo)明確,不需特殊設(shè)備,造模主要著眼于直接造成外周血中的細(xì)胞數(shù)量減少,接近臨床中醫(yī)血虛證。其發(fā)生發(fā)展的分子機(jī)制為:骨髓細(xì)胞凋亡,細(xì)胞周期紊亂,結(jié)果造成骨髓中干祖細(xì)胞的能力降低,進(jìn)而引起外周血象下降。四物湯防治小鼠血虛證的機(jī)理為:四物湯提前給藥可預(yù)防綜合放血引起的凋亡,增強(qiáng)免疫力,四物湯治療給藥可促進(jìn)骨髓細(xì)胞增殖能力,增加外周血中的紅細(xì)胞和血紅蛋白。
[Abstract]:Blood deficiency is a common clinical syndrome, but the establishment of pharmacology of blood deficiency model close to the TCM clinical literature is rarely reported. This topic is in the (60) ~Co gamma ray irradiation and cyclophosphamide intraperitoneal injection production based on the mouse model of blood deficiency, chronic bleeding, food restriction, swimming has set up a comprehensive phlebotomy blood deficiency model, and separately from the peripheral blood, cell cycle, (CD34) ~ + cells, hematopoietic progenitor cell colony formation ability, bone marrow and spleen cell apoptosis, discusses the mechanism of phlebotomy blood deficiency of the occurrence and development, in order to improve the experimental data, different methods of establishing the model of blood deficiency and overall. Changes of blood deficiency model objective evaluation of traditional Chinese medicine. Siwu Decoction is a famous prescription of a blood circulation by regulating menstruation, prepared rehmannia root, angelica, Radix Paeoniae Alba, Rhizoma Chuanxiong, has a definite effect. This study systematically The effect of Siwu Decoction on the model mice of blood deficiency syndrome was observed, and the molecular mechanism of the effect of Siwu Decoction on the blood deficiency syndrome model of the comprehensive blood letting method was clarified.
This study through the daily press 75g.kg~ (-1) weight restriction, then 20min and 0.4ml to establish a comprehensive swimming bloodletting bloodletting model of blood deficiency mice model, 15d. experimental results showed that the mice in the model of dry fur, 9D low gloss, lazy and so on the different degree of blood deficiency; peripheral blood count red blood cell and hemoglobin from the model until the date of the 14d at the end of the modeling are decreased significantly; the proportion of (CD34) ~ + cells in the 7,12d group was significantly higher than that of normal bone marrow cells; the number of G0/1 cells in the 4,7,21d was significantly lower than the normal group; while the number of S cells and the proliferation index in the 4,7,21d significantly higher than the normal group; the thymus and thymus index decreased significantly.
To investigate the mechanism of Siwu Decoction in prevention and treatment of comprehensive blood letting of blood deficiency, were given by gavage in mice before and after making models. The results showed that: Siwu Decoction treatment for bone marrow cell proliferation; apoptosis of bone marrow hematopoietic stem progenitor cells; enhance the immune function of blood deficiency mice. Siwu decoction for prevention the role of the drug is more obvious on mouse bone marrow hematopoietic stem cells, peripheral blood cells can obviously promote the recovery of bone marrow cells; reduce G2/M block, protect the bone marrow and bone marrow cells into effect S phase from G1 phase. Bone marrow hematopoietic stem progenitor cell proliferation, inhibition of apoptosis of bone marrow hematopoietic stem progenitor cells may be one of the mechanisms of comprehensive phlebotomy blood deficiency treatment of Siwu Decoction.
Comprehensive phlebotomy, (60) ~Co gamma ray irradiation, blood deficiency model by intraperitoneal injection of cyclophosphamide to establish the comparative experimental results show that the comprehensive bloodletting group of red blood cell and hemoglobin decreased significantly, (60) ~Co gamma ray irradiation group of white blood cells decreased significantly, cyclophosphamide group white cells and red blood cells decreased significantly; comprehensive bleeding group in the modeling of the 15d (60) ~Co gamma ray irradiation group after modeling 7d, cyclophosphamide group at CFU-E, 7d after modeling BFU-E, CFU-GM were significantly reduced; comprehensive bleeding group (CD34) ~ + cells increased significantly in 8,11d (60) ~Co gamma ray irradiation of CD34+ cells from the model group after 14d were significantly lower than normal group, cyclophosphamide group from after the model first increased and then decreased; three models of thymus index were significantly lower than normal group, but the comprehensive index of spleen bleeding group was significantly higher than the normal group and the irradiation group instead.
In general, the comprehensive index of phlebotomy is clear, no need of special equipment, molding mainly focus on directly caused the number of cells in peripheral blood decreased, close to the clinical medicine of blood deficiency. The molecular mechanism of the occurrence and development of bone marrow cells: apoptosis, cell cycle disorder, resulting in reduced ability of stem and progenitor cells in the bone marrow. Which causes the decrease of peripheral blood. The mechanism of Siwu Decoction on mice model of blood deficiency syndrome: apoptosis, early administration of Siwu Decoction can prevent bleeding caused by comprehensive enhance immunity, Siwu Decoction treatment can promote the proliferation of bone marrow cells, red blood cells increased in the peripheral blood and hemoglobin.
