球形幽門螺桿菌對胃上皮細胞增殖與凋亡及細胞內轉錄因子Egr-1表達的影響

【摘要】: 幽門螺桿菌(Helicobacter pylori, Hp)在營養(yǎng)不足,抗生素處理等脅迫因素作用下,其形態(tài)會發(fā)生適應性轉變,由螺旋形轉變?yōu)榍蛐。球形幽門螺桿菌具有免疫原性低,不易清除等特點。研究球形幽門螺桿菌對宿主細胞增殖與凋亡的影響,以及對宿主細胞內轉錄因子Egr-1表達的影響,將有助于了解球形Hp感染與各種胃病特別是胃癌發(fā)生的關系,從細胞微生物學的角度深入探討幽門螺桿菌的致病機理。 一、球形Hp對胃上皮細胞增殖與凋亡的影響 目的:了解球形幽門螺桿菌對人胃上皮細胞增殖與凋亡的影響。方法:選擇正常胃上皮細胞株GES-1和胃癌細胞株AGS為研究對象,將球形和螺旋形Hp分別與它們共培養(yǎng)。利用CCK-8法檢測活細胞的細胞增殖情況,用TUNEL法檢測細胞凋亡情況。并比較在相同感染密度下,兩種形態(tài)的Hp對胃上皮細胞的增殖與凋亡的作用強度是否存在差異。結果:1.在低感染密度狀態(tài)下,球形Hp的促細胞增殖作用高于對照組(PI=2.6%~5.0%),P0.05。當MOI=32:1時,球形Hp處理AGS細胞OD值為2.902±0.072,細胞增殖率(PI)62.58%。明顯高于螺旋形Hp處理組(PI=18.04%),P0.05。當MOI=16:1時,球形Hp處理GES-1細胞OD值為1.808±0.156,細胞增殖率32.07%,明顯高于螺旋形Hp處理組(PI=21.91%),P0.05。2.在低感染密度狀態(tài)下,球形Hp的誘導凋亡作用較弱,與對照組(AI=3.24%~5.42%)相比,無明顯差異,P0.05。當MOI=32:1時,球形Hp處理的AGS細胞,凋亡率(AI)為5.94%,明顯低于螺旋形Hp處理組(AI=14.52%),P0.05。當MOI=16:1時,球形Hp處理GES-1細胞,凋亡率為6.81%,明顯低于螺旋形Hp處理組(AI=16.25%),P0.05。結論:1.在Hp的低感染密度狀態(tài)下,與螺旋形Hp相比,球形Hp對胃上皮細胞表現出較強的促增殖作用和相對較弱的誘導凋亡的作用。故球形Hp對體外胃上皮細胞具有較強的促增殖效應。2.球形Hp對正常胃上皮細胞和胃癌細胞增殖與凋亡的影響基本一致。 二、球形Hp刺激AGS細胞增殖過程中轉錄因子Egr-1的作用 目的:活化的Egr-1在胃上皮細胞的惡性轉化中具有重要作用。檢測球形Hp對胃
[Abstract]:The morphology of Helicobacter pylori (HP) changed from spiral to spherical under stress factors such as inadequate nutrition and antibiotic treatment.Helicobacter pylori globular has low immunogenicity and is difficult to be cleared.To study the effects of Helicobacter pylori on the proliferation and apoptosis of host cells and on the expression of transcription factor Egr-1 in host cells will contribute to the understanding of the relationship between spherical HP infection and the occurrence of various gastric diseases, especially gastric cancer.To explore the pathogenic mechanism of Helicobacter pylori from the point of view of cell microbiology.1. Effects of spherical HP on proliferation and apoptosis of gastric epithelial cellsObjective: to investigate the effect of Helicobacter pylori on proliferation and apoptosis of human gastric epithelial cells.Methods: normal gastric epithelial cell line (GES-1) and gastric cancer cell line (AGS) were selected and cocultured with spherical and spiral HP respectively.CCK-8 assay was used to detect cell proliferation and TUNEL assay was used to detect cell apoptosis.At the same infection density, the effects of HP on gastric epithelial cell proliferation and apoptosis were compared.The result is 1: 1.Under the condition of low infection density, the proliferative effect of globular HP was higher than that of the control group (PI2.6and 5.0P0.05).When treated with MOI=32:1, the OD value of AGS cells treated with spherical HP was 2.902 鹵0.072, and the cell proliferation rate was 62.58.It was significantly higher than that of helical HP treatment group (P 0.05).When treated with MOI=16:1, the OD value of GES-1 cells treated with spherical HP was 1.808 鹵0.156, and the cell proliferation rate was 32.07, which was significantly higher than that of helical HP treatment group (P _ I _ (21.91)) and P _ (0.05.2).Under the condition of low infection density, the apoptosis-inducing effect of globular HP was weaker than that of the control group (AI3.24 and 5.42), there was no significant difference between them (P0.05).When treated with MOI=32:1, the apoptotic rate of AGS cells treated with spherical HP was 5.94%, which was significantly lower than that of spiral HP treatment group (14.52% P0.05).When treated with MOI=16:1, the apoptotic rate of GES-1 cells treated with spherical HP was 6.81, which was significantly lower than that of helical HP treatment group (P 0.05).Conclusion 1.Under the condition of low infection density of HP, compared with helical HP, spherical HP could promote the proliferation and induce apoptosis of gastric epithelial cells.Therefore, spherical HP has a strong proliferative effect on gastric epithelial cells in vitro.The effects of spherical HP on the proliferation and apoptosis of normal gastric epithelial cells and gastric cancer cells were consistent.Second, the role of transcription factor Egr-1 in the proliferation of AGS cells stimulated by spherical HPObjective: activated Egr-1 plays an important role in malignant transformation of gastric epithelial cells.Detection of spherical HP on stomach
