【摘要】：[研究背景]ndrg2與ndrg1、ndrg3和ndrg4共同構(gòu)成了一個新的基因家族，即ndrg家族。該基因家族參與了細胞的生長，但具體機制仍不清楚。人ndrg2、鼠ndrg2分別由我導師鄧艷春教授和日本研究小組發(fā)現(xiàn)，氨基酸序列分析顯示，，人ndrg2、鼠ndrg2的同源性高達95％。人ndrg2基因的基因組全長為9.33kb，位于染色體的14q11.2，mRNA全長2024bp編碼含357個氨基酸殘基、分子量約為40.79kD的蛋白質(zhì)。曾命名為p40。同源性分析表明，其與ndrg1(也稱做腦發(fā)育相關(guān)蛋白)具有高度同源性，并含有一個酰基攜帶蛋白樣結(jié)構(gòu)域，故稱為ndrg2。研究結(jié)果初步顯示，ndrg2在多種正常組織中高表達，而在某些腫瘤組織和腫瘤細胞系中低表達或無表達，在體外可抑制多種腫瘤細胞系的生長(如膠質(zhì)瘤細胞系BT 325)，推測該基因可能與腫瘤的發(fā)生發(fā)展有關(guān)，因此提出ndrg2可能為新的候選抑癌基因。新近資料表明，ndrg2參與了抑郁癥和阿爾采默病等腦部疾病的發(fā)生發(fā)展，但這個基因與腦功能及神經(jīng)元發(fā)育、成熟有何關(guān)系，目前尚不清楚。[研究目的]該實驗旨在系統(tǒng)分析ndrg2在胎腦發(fā)育不同階段表達分布的時空特點，以期弄清ndrg2在胎腦各腦功能區(qū)的表達特點及在胎腦成熟過程中的表達趨勢，為進一步研究其功能提供實驗依據(jù)。[研究方法]1．用免疫組化染色方法從蛋白質(zhì)水平確定ndrg2在16～28周胎腦各腦功能區(qū)的時空表達特點及表達趨勢。用抗ndrg2單克隆抗體，對16～28周各月份各腦功能區(qū)石蠟切片進
[Abstract]:Background ndrg2, together with ndrg1ndrg3 and ndrg4, constitute a new gene family, ndrg family.The gene family is involved in cell growth, but the exact mechanism remains unclear.The amino acid sequence analysis of human ndrg2 and mouse ndrg2 showed that the homology of human ndrg2 and mouse ndrg2 was 95%.The full length of human ndrg2 gene is 9.33 kb. The full-length 2024bp of 14q11.2mRNAs encodes a protein with a molecular weight of about 40.79kD containing 357 amino acid residues.Once named p40.Homology analysis showed that it had high homology with ndrg1 (also known as brain development-related protein) and contained an acyl-carrying protein-like domain, so it was called ndrg2.The results showed that the expression of ndrg2 was high in many normal tissues, but low or no expression in some tumor tissues and tumor cell lines.Recent data indicate that ndrg2 is involved in the development of brain diseases such as depression and Alzheimer's disease, but it is not clear how this gene is related to brain function and neuronal development and maturation.[objective] to systematically analyze the temporal and spatial characteristics of expression and distribution of ndrg2 in different stages of fetal brain development in order to find out the expression characteristics of ndrg2 in various functional areas of fetal brain and its expression trend during fetal brain maturation.To provide experimental basis for further study of its function.[research methods] 1.Immunohistochemical staining was used to determine the temporal and spatial expression characteristics and expression trend of ndrg2 in various functional brain regions of the fetal brain at 1628 weeks.Using anti ndrg2 monoclonal antibody, paraffin sections of the functional regions of the brain in each month of 16 weeks 28 weeks were embedded in paraffin sections.
