陰虛熱盛型2型糖尿病動(dòng)物模型的初步研究

【摘要】:目的:建立陰虛熱盛型2.型糖尿病的病癥結(jié)合動(dòng)物模型,探討陰虛熱盛型2型糖尿病動(dòng)物模型的成模標(biāo)準(zhǔn)。 方法:將80只清潔級(jí)WISTAR大鼠隨機(jī)分為正常對(duì)照組、高脂對(duì)照組、糖尿病空白對(duì)照組和陰虛熱盛模型組4組。對(duì)模型組大鼠灌服中藥干姜、附子,其余各組大鼠不做任何處理,連續(xù)4周,然后用玉女煎對(duì)模型組大鼠進(jìn)行反證。 結(jié)果: 1.中藥造模后大鼠出現(xiàn)多飲、多食、多尿、身體消瘦、大便干燥、灌藥時(shí)掙扎明顯。舌暗紅、少苔等征象。 2.客觀指標(biāo):觀察了血糖、血脂、皮質(zhì)醇、胰島素水平、甲狀腺功能、血栓素B_2、6-Keto-PGF_(1a)水平: 模型組血糖顯著升高,與正常對(duì)照組比較有明顯差異,與糖尿病空白對(duì)照組無(wú)明顯差異。 模型組血清甘油三酯、總膽固醇和低密度脂蛋白均較正常對(duì)照組高,有顯著性差異。與糖尿病空白對(duì)照組比較無(wú)顯著性差異。 模型組血漿皮質(zhì)醇水平明顯高于正常對(duì)照組有顯著性差異;與糖尿病空白對(duì)照組無(wú)明顯差異。 模型組胰島素水平較正常對(duì)照組降低,與正常對(duì)照組有顯著性差異;與糖尿病空白對(duì)照組無(wú)明顯性差異。 模型組大鼠FT_3升高,與各對(duì)照組有顯著性差異。 模型組大鼠和糖尿病空白對(duì)照組血栓素B2水平升高,于正常對(duì)照組有明顯差異;模型組大鼠和糖尿病空白對(duì)照組6-Keto-PGF_(1a)水平降低,,與正常對(duì)照組有明顯性差異;但模型組與糖尿病空白對(duì)照組無(wú)顯著性差異。 用玉女煎反證后以上指標(biāo)有所恢復(fù),其中甘油三酯、總膽固醇、血栓素B_2水平與反證前有顯著性差異,接近糖尿病空白對(duì)照組。 結(jié)論: 在高脂加鏈脲佐菌素誘發(fā)糖尿病造模方法的基礎(chǔ)上,再根據(jù)中醫(yī)理論運(yùn)用中藥四氣五味的藥性來(lái)研制2型糖尿病的中醫(yī)證候的動(dòng)物模型是切實(shí)可行的。從而將陰虛熱盛“證”動(dòng)物模型與糖尿“病”動(dòng)物模型結(jié)合起來(lái),使糖尿病動(dòng)物模型證候化,與中醫(yī)臨床辨證論治相一致,以便為廣泛開(kāi)展糖尿病的研究提供良好的動(dòng)物模型。
[Abstract]:Objective: to establish an animal model of type 2 diabetes mellitus with deficiency of yin and heat. Methods: eighty clean grade WISTAR rats were randomly divided into 4 groups: normal control group, hyperlipidemia control group, diabetes blank control group and Yin deficiency heat Sheng model group. After 4 weeks, the rats in the model group were treated with Yusu decoction. Results:. 1. After making model of traditional Chinese medicine, the rats showed signs of polydrink, polyfood, polyuria, thin body, dry stool, obvious struggle when perfusing medicine, dark red tongue, less fur and so on. 2. Objective measures: blood glucose, blood lipids, cortisol, insulin level, thyroid function, thromboxane B _ (26-Keto-PGF) levels were observed:. The blood glucose in the model group was significantly higher than that in the normal control group, but there was no significant difference between the model group and the diabetic blank control group. Serum triglyceride, total cholesterol and low density lipoprotein in the model group were significantly higher than those in the normal control group, but there was no significant difference between the model group and the control group. The plasma cortisol level in the model group was significantly higher than that in the normal control group, but there was no significant difference between the model group and the diabetic blank control group. The insulin level in the model group was lower than that in the normal control group, and there was a significant difference between the model group and the normal control group, but there was no significant difference between the model group and the diabetic blank control group. FT_3 in the model group was significantly higher than that in the control group. The level of thromboxane B2 in the model group and the diabetic blank control group was significantly higher than that in the normal control group, and the level of 6-Keto-PGFS-1 a in the model group and the diabetic blank control group was significantly lower than that in the normal control group. However, there was no significant difference between the model group and the diabetic blank control group. The levels of triglyceride, total cholesterol and thromboxane B _ 2 were significantly different from those before reverse, which were close to the control group of diabetes mellitus. Conclusion:. Based on the method of diabetes induced by hyperlipidemia and streptozotocin, According to the theory of traditional Chinese medicine, it is feasible to develop the animal model of TCM syndromes of type 2 diabetes mellitus by using the medicine of four qi and five flavors of traditional Chinese medicine, so as to combine the animal model of "syndrome" of Yin deficiency and heat with the animal model of "disease" of sugar and urine. The syndromes of diabetic animal model are consistent with TCM clinical syndrome differentiation and treatment, so as to provide a good animal model for the extensive research of diabetes mellitus.
