大鼠腦紋狀體邊緣區(qū)多巴胺受體及其與學(xué)習(xí)記憶功能的研究

【摘要】:腦紋狀體邊緣區(qū)(marginal division,MrD)是舒斯云等1987年在研究大鼠腦紋狀體時發(fā)現(xiàn)的位于新紋狀體尾內(nèi)側(cè)、環(huán)繞蒼白球頭外側(cè)的一個由梭形細(xì)胞構(gòu)成的新亞區(qū),隨后在貓、猴和人腦內(nèi)也發(fā)現(xiàn)存在類似的結(jié)構(gòu),其富含多種神經(jīng)肽類陽性纖維、終末和胞體,與腦內(nèi)包括海馬、杏仁核、Meynert氏基核和前額葉皮質(zhì)等多個學(xué)習(xí)記憶相關(guān)腦區(qū)存在著結(jié)構(gòu)和/或功能上的聯(lián)系,是一個在細(xì)胞形態(tài)、神經(jīng)遞質(zhì)和受體等生物大分子的分布,以及纖維聯(lián)系方面均有別于紋狀體其他部位的獨(dú)特的亞區(qū)。該區(qū)富含SP、5-HT、DA、L-ENK、NT、SOM和TH等多種神經(jīng)肽陽性纖維和胞體。該區(qū)和Meynert基底核及黑質(zhì)等部位均有纖維聯(lián)系。而且,己證實(shí)大鼠的MrD參與了電Y迷宮厭暗逃避條件反射聯(lián)合型學(xué)習(xí),在相同的行為模式訓(xùn)練后可觀察到大鼠MrD細(xì)胞有c-fos、c-jun即早基因和與長期記憶形成密切相關(guān)的細(xì)胞核轉(zhuǎn)錄調(diào)節(jié)因子pCREB表達(dá)。最近,又進(jìn)一步發(fā)現(xiàn)MrD涉及人的聽覺數(shù)字工作記憶,臨床上也觀察到因MrD受損而出現(xiàn)認(rèn)知功能障礙的患者。 多巴胺(DA)是腦中豐富的兒茶酚胺類神經(jīng)遞質(zhì),廣泛參與運(yùn)動、認(rèn)知、學(xué)習(xí)與記憶、情感、內(nèi)分泌、正性強(qiáng)化等多種生理功能的調(diào)節(jié)。帕金森氏病、精神分裂癥、抽動穢語綜合征(Tourette's syndrome)、藥物成癮、高泌乳素血癥等疾病都與多巴胺遞質(zhì)傳遞障礙有關(guān)。多巴胺發(fā)揮生理、病理等方面的作用是
