用hTSHR活化的脾細(xì)胞免疫小鼠建立甲狀腺相關(guān)性眼病模型的研究

【摘要】:目的 用 hTSHR 活化的脾細(xì)胞免疫同源 BALB/c 小鼠建立甲狀腺相關(guān)性眼病(TAO)動(dòng)物模型。 方法 將 6-8 周雌性 BALB/c 小鼠預(yù)處理后隨機(jī)分為兩組(a 組和b 組),分別用重組質(zhì)粒 pcDNA3.1/hTSHR 和空質(zhì)粒 pcDNA3.1 免疫兩組小鼠,并在免疫后的第 3 周、第 6 周加強(qiáng)免疫兩次。18 周后分別處死兩組小鼠,取眼眶組織做病理學(xué)觀察,并機(jī)械分離脾細(xì)胞。將 6-8 周雌性 BALB/c 小鼠隨機(jī)分為兩組(c 組和 d 組),分別接受 a 組和 b 組小鼠的脾細(xì)胞免疫。4 周后處死小鼠,取眼眶組織做病理學(xué)觀察,用放射免疫法測(cè)定血清 TT4 水平、免疫放射法測(cè)定血清 TSH 水平,ELISA 法檢測(cè)血清 TRAb。 結(jié)果 a 組(重組質(zhì)粒免疫組):約 25%的小鼠眼眶組織出現(xiàn)粘液變性、水腫,但無(wú)明顯纖維組織及脂肪組織增生,肌纖維無(wú)變性斷裂。c 組(活化脾細(xì)胞免疫組):約有 50%的小鼠眼眶組織出現(xiàn)明顯粘液變性、水腫,局部纖維組織和脂肪組織增生,肌纖維透明變性斷裂;電鏡下可見滑面內(nèi)質(zhì)網(wǎng)擴(kuò)張,肌原纖維排列紊亂。b 組(空質(zhì)粒免疫組)和 d 組(未活化脾細(xì)胞免疫組):小鼠的眼眶組織均無(wú)形態(tài)學(xué)改變。脾細(xì)胞免疫后 4 周,血清 TT4:c 組(7.130±1.017 ug/dl)與 d 組(6.431±0.573 ug/dl)相比,TT4 水平顯著升高(p=0.036)。血清 TSH:c組(0.070±0.032 uIU/ml)與 d 組(0.098±0.020 uIU/ml)相比,TSH 水平顯著降低(p=0.020)。其中,2 只小鼠的血清 TT4 高于參考范圍上限,血清 TSH 低于參考范圍下限,符合甲狀腺功能亢進(jìn)的血清學(xué)
[Abstract]:Objective to establish an animal model of thyroid associated ophthalmopathy (TAO) in BALB/c mice immunized with spleen cells activated by hTSHR. Methods female BALB/c mice of 6-8 weeks were randomly divided into two groups: group A and group b. The mice were immunized with recombinant plasmid pcDNA3.1/hTSHR and empty plasmid pcDNA3.1 at the third week after immunization. After 6 weeks, the mice were killed and the orbital tissues were taken for pathological observation. Six to eight weeks old female BALB/c mice were randomly divided into two groups: group C and group d. The mice of group a and group b were immunized with spleen cells for 4 weeks, and then the mice were killed, and the orbital tissues were taken for pathological observation. Serum TT4 levels were measured by radioimmunoassay and serum TSH levels by immunoradiometric assay. Results in group a (recombinant plasmid immunized group), about 25% mice had mucus degeneration and edema, but there was no obvious proliferation of fibrous tissue and adipose tissue. In group c (activated splenocyte immunity group), about 50% of mouse orbital tissues showed obvious mucus degeneration, edema, proliferation of local fibrous tissue and adipose tissue, and hyaline degeneration of muscle fiber. Electron microscope showed smooth endoplasmic reticulum dilatation, myofibril arrangement disorder. Group b (empty plasmid immunized group) and group d (unactivated splenocyte immunization group): no morphological changes were found in the orbital tissues of mice. The spleen cells were immunized 4 weeks after immunization. Compared with d group (6.431 鹵0.573 ugr / dl), the level of TT4 in serum TT4:c group was significantly higher than that in group d (0.070 鹵0.032 渭 mol 路dll), and the level of TT4 in serum TSH:c group was significantly lower than that in group d (0.098 鹵0.020 uIU-dll). The serum TT4 of 2 mice was higher than the upper limit of reference range. Serum TSH was lower than the lower limit of reference range, which was in accordance with the serology of hyperthyroidism.
