HCK與IFRD1基因在胚胎心臟中時空表達的研究

【摘要】: 【摘要】 目的檢測HCK(hemopoietic cell kinase)基因在大鼠胚胎心臟發(fā)育過程中mRNA表達的變化,分析其與心臟發(fā)育的關系。方法取大鼠胚胎12、15、19d心臟,采用RT-PCR技術檢測心臟組織中HCK基因mRNA的表達豐度,并應用原位雜交技術對HCK基因表達進行定位分析。結果在胚胎12、15、19d心臟中,HCK基因均有表達,但主要表達于心臟間質,在心肌細胞中不表達,其表達豐度分別為0.19±0.09、0.38±0.07、0.59±0.10,隨胚齡增加呈逐漸上調趨勢(P<0.05)。結論HCK基因表達于心臟間質,在心肌細胞中無表達,其表達豐度與大鼠胚胎心臟發(fā)育時間有關,可能參與心臟發(fā)育。 目的檢測大鼠胚胎心臟發(fā)育過程中IFRD1(interferon-related developmental regulator 1)基因mRNA與蛋白的表達變化。方法取大鼠胚胎12、15、19d心臟,應用RT-PCR、Western blot、免疫組化技術,檢測IFRD1基因mRNA與蛋白的表達。結果在大鼠胚胎12、15、19d天心臟中,IFRD1基因mRNA與蛋白均有表達,且呈逐漸下調趨勢;其蛋白主要表達于心房、心室、肌小梁等心肌組織中,而在心內膜墊、瓣膜、流出道、大血管內層等處均未見表達。結論IFRD1基因隨大鼠胚胎心臟發(fā)育表達逐漸下調,特異表達于心房、心室、肌小梁等大鼠胚胎心臟組織中,可能參與心臟發(fā)育。
[Abstract]:[abstract]. Objective to detect the expression of HCK(hemopoietic cell kinase gene in rat embryonic heart and to analyze the relationship between mRNA expression and cardiac development. Methods the expression of HCK gene mRNA was detected by RT-PCR technique. In situ hybridization technique was used to localize the expression of HCK gene. Results the expression of HCK gene was found in the heart of 1215 ~ 19d embryo, but mainly in the interstitial of the heart, but not in the cardiomyocytes. The expression abundance of HCK was 0.19 鹵0.09 鹵0.38 鹵0.070.59 鹵0.10, which was gradually up-regulated with the increase of embryonic age (P < 0.05). Conclusion the expression of HCK gene is expressed in the interstitial of the heart, but not in the cardiomyocytes, and its expression abundance is related to the development time of rat embryonic heart and may participate in the development of the heart. Objective to detect the expression of IFRD1(interferon-related developmental regulator 1 (IFRD1(interferon-related developmental regulator 1) gene mRNA and protein during the development of rat embryonic heart. Results the expression of mRNA and protein of IFRD1 gene was down-regulated in the rat heart on day 1215 and 19. The protein was mainly expressed in myocardium such as atrium, ventricle and trabecula, but in endocardium. Conclusion the expression of IFRD1 gene is down-regulated gradually with the development of rat embryonic heart, and is specifically expressed in rat embryonic heart tissues such as atrium, ventricle and trabecula, which may be involved in cardiac development.
