【摘要】：目的了解吉氏染色法對瘧原蟲染色效果的影響因素,尋找出最佳的染色條件。方法比較吉氏染色法不同染色時間、不同染液濃度以及染液中顆粒性雜質(zhì)的去除對外周血中瘧原蟲的染色情況,尋找最佳效果。結(jié)果在本實(shí)驗(yàn)室自制吉氏染液薄血膜染色的最佳染色時間和最佳染色濃度分別為10min、1∶5稀釋;15min、1∶10稀釋;20min、1∶15稀釋以及25min、1∶20稀釋;厚血膜染色5min就能達(dá)到最佳結(jié)果。自制吉氏原液經(jīng)離心后再稀釋染色,染色效果比未離心的原液好,鏡下觀察雜質(zhì)少而干凈。結(jié)論吉氏染色法對瘧原蟲的染色效果的影響因素有染色時間和染液濃度等,相關(guān)實(shí)驗(yàn)室應(yīng)予關(guān)注。
[Abstract]:Objective to investigate the factors influencing the staining effect of Plasmodium falciparum and find out the best staining conditions. Staining of Plasmodium falciparum in peripheral blood with different concentrations of dye solution and removal of particulate impurities in the solution, Results in our laboratory, the best dyeing time and the best dyeing concentration of the thin blood membrane dyeing solution were 10 min / 1: 5, 15 min ~ (-1), 1: 10, 20 min ~ (-1) 1: 15 and 25 min ~ (-1): 20% ~ 20%, respectively, and the best dyeing time and the best dyeing concentration were 10 min / 1: 5 and 15 min / 10 respectively. The best result can be obtained by thick blood membrane staining with 5min. After centrifugation and dilution dyeing, the dyeing effect is better than that without centrifugation. Conclusion the influence factors of the staining effect of Plasmodium falciparum are the time of staining and the concentration of dye solution, which should be paid more attention to in the laboratory.
