人胎兒骨髓間充質(zhì)干細(xì)胞的生物學(xué)特性研究

【摘要】: 間充質(zhì)干細(xì)胞(MSC)是一種具有多向分化功能的細(xì)胞群體,早期主要從成人骨髓中分離獲得,近年研究表明,多種組織中均含有MSC如外周血、臍帶血、脂肪組織、羊水以及其他的一些胎兒組織。MSC在體外能夠擴(kuò)增,其在適合的條件下,可向包括脂肪細(xì)胞、成骨細(xì)胞、軟骨細(xì)胞、心肌細(xì)胞、神經(jīng)細(xì)胞、肝細(xì)胞的多種組織細(xì)胞分化。MSC具有免疫調(diào)節(jié)作用,MSC誘導(dǎo)淋巴細(xì)胞通過分泌可溶性細(xì)胞因子而產(chǎn)生免疫抑制效應(yīng),在混合淋巴細(xì)胞培養(yǎng)中能抑制T細(xì)胞的增殖,與造血干細(xì)胞共輸可降低移植物抗宿主病的發(fā)生率。最近人們發(fā)現(xiàn)它能夠逆轉(zhuǎn)內(nèi)臟和肝臟的4期急性GVHD。鑒此MSCs作為造血重建的促進(jìn)劑,預(yù)防和治療GVHD,它們在器官移植排斥的治療和自身免疫性疾病的組織修復(fù)中具有潛在的應(yīng)用價(jià)值。 本實(shí)驗(yàn)應(yīng)用細(xì)胞生物學(xué)和免疫學(xué)等多種技術(shù): (1)分離培養(yǎng)并鑒定人胎兒源骨髓間充質(zhì)干樣細(xì)胞(FMSCs) ;(2)觀察不同劑量的射線輻照對增殖;細(xì)胞周期的影響,比較其在放射損傷前后的生物學(xué)特性,輻照后成脂、成骨分化的潛能;(3)研究人胎兒源骨髓間充質(zhì)干細(xì)胞在免疫調(diào)節(jié)中的作用。 目的:分離培養(yǎng)并鑒定人胎兒源骨髓間充質(zhì)樣干細(xì)胞(FMSCs) ;比
[Abstract]:MSC (Mesenchymal Stem Cell) is a multifunctional cell population, which was isolated from adult bone marrow in the early stage. Recent studies have shown that many tissues contain MSC, such as peripheral blood, umbilical cord blood, adipose tissue. Amniotic fluid and some other fetal tissues, such as. MSC, can be expanded in vitro to include adipocytes, osteoblasts, chondrocytes, cardiomyocytes, nerve cells, and, where appropriate, adipocytes, osteoblasts, chondrocytes, cardiomyocytes, nerve cells. MSC can induce lymphocytes to produce immunosuppressive effect by secreting soluble cytokines, which can inhibit the proliferation of T cells in mixed lymphocyte culture. Coadministration with hematopoietic stem cells can reduce the incidence of graft-versus-host disease. It has recently been found to be able to reverse phase 4 acute GVHDs of viscera and liver. This MSCs is used as an accelerator for hematopoietic reconstitution. The prevention and treatment of GVHD, which have potential application value in the treatment of organ transplantation rejection and tissue repair of autoimmune diseases. In this study, cell biology and immunology were used to observe the effects of different doses of radiation irradiation on proliferation and cell cycle of human fetal bone marrow mesenchymal mesenchymal stem cells (FMSCs2). To study the role of human fetal bone marrow mesenchymal stem cells in immunomodulation by comparing their biological characteristics before and after radiation injury and the potential of adipogenesis and osteogenic differentiation after irradiation. Objective: to isolate and identify human fetal bone marrow mesenchymal stem cells (MSCs).
