建立兔腰椎間盤退變模型的實(shí)驗(yàn)研究

【摘要】: 【目的】:探討纖維環(huán)刺傷法誘導(dǎo)兔腰椎間盤退變建立動(dòng)物模型的可行性,建立一種緩慢進(jìn)展、可重復(fù)性好的兔腰椎間盤退變的動(dòng)物模型,適用于研究椎間盤退變的病因?qū)W、病理生理學(xué),適用于檢驗(yàn)可靠、有效的治療椎間盤退變的新方法,例如生長(zhǎng)因子療法、基因療法、細(xì)胞療法、組織工程等。 【方法】:24只普通級(jí)雄性新西蘭大白兔(重量約3.0kg,1歲齡),手術(shù)取右側(cè)腹膜后入路,用16號(hào)穿刺針于L2/3椎間盤纖維環(huán)右前外側(cè)刺入,深度為5mm,分別于術(shù)前及術(shù)后4、8、16周對(duì)造模的椎間盤(L2/3)及正常椎間盤(L4/5)進(jìn)行磁共振(MRI)檢查,動(dòng)物處死取材后行組織學(xué)及免疫組化觀察。 【結(jié)果】:①術(shù)前到術(shù)后16周組,造模組椎間盤毮核MRI T2W1信號(hào)呈逐漸減弱趨勢(shì)(P0.01);②組織學(xué)觀察到毮核脊索細(xì)胞逐漸減少,并逐漸被纖維軟骨組織替代,纖維環(huán)與毮核之間失去明顯的界限;③免疫組化觀察到毮核中II型膠原表達(dá)較對(duì)照組逐漸減少(P0.01),而I型膠原表達(dá)較對(duì)照組逐漸增多(P0.01)。 【結(jié)論】:1.本實(shí)驗(yàn)通過MRI影像學(xué)分析、組織學(xué)觀察及免疫組化觀察證實(shí)了兔椎間盤退變的基本特征;2.纖維環(huán)刺傷法可以誘導(dǎo)兔椎間盤緩慢退變,為椎間盤退變的進(jìn)一步研究提供動(dòng)物模型。
[Abstract]:[objective]: to investigate the feasibility of rabbit lumbar disc degeneration induced by fibrous ring puncture, and to establish a slowly progressive and reproducible animal model of lumbar disc degeneration, which is suitable for the study of the etiology of intervertebral disc degeneration. Pathophysiology is suitable for testing reliable and effective new methods for the treatment of intervertebral disc degeneration, such as growth factor therapy, gene therapy, cell therapy, tissue engineering and so on. [methods] Twenty-four normal male New Zealand white rabbits (weighing about 3.0 kg / 1 years old) were operated on the right retroperitoneal approach and punctured on the right anterolateral side of the L 2 / 3 intervertebral disc fiber ring with 16 puncture needle. The depth was 5 mm. MRI was performed on L2 / 3 of model intervertebral disc and L4 / 5 of normal intervertebral disc before operation and 8 weeks after operation. The animals were sacrificed for histological and immunohistochemical observation. [results] the signal intensity of MRI T2W1 in the intervertebral disc nucleus of the model group decreased gradually from pre-operation to 16th week after operation. Histologically, the notochordal cells of the nucleus carinii were gradually decreased and replaced by fibrochondrocytes. The expression of type II collagen was decreased gradually compared with the control group, while the expression of type I collagen was gradually increased than that of the control group. [conclusion] 1. The basic features of disc degeneration in rabbits were confirmed by MRI imaging analysis, histological observation and immunohistochemical observation. 2. The method of fibrous ring puncture can induce slow degeneration of intervertebral disc in rabbits. To provide animal models for further study of disc degeneration.
