輻照結合束縛應激致血虛肝郁大鼠證候模型的建立

【摘要】:目的:建立輻照結合束縛應激致血虛肝郁證大鼠模型。方法:SD雄性大鼠隨機分為空白組、輻照組、束縛組、模型組,模型組采用大鼠單籠飼養(yǎng)、輻照結合束縛21d造模,檢測各組大鼠體質量、外周血象并進行糖水實驗、敞箱實驗和跳臺實驗,測定血漿中促腎上腺皮質激素(ACTH)、皮質酮(CORT),下丘腦促腎上腺皮質激素釋放激素(CRH)和大腦海馬單胺類神經遞質5-羥色胺(5-HT)和多巴胺(DA)等含量。結果:模型組大鼠生長緩慢,外周白細胞,紅細胞、血紅蛋白下降明顯(P0.05),血漿中ACTH明顯升高(P0.05),CORT和下丘腦CRH含量有升高趨勢,腦內單胺類神經遞質5-HT和DA含量明顯降低(P0.05)。結論:從血象、行為學、下丘腦-垂體-腎上腺(HPA)軸和腦內單胺類神經遞質等方面驗證了輻照結合束縛應激造成的大鼠模型可作為中醫(yī)血虛肝郁證的動物模型。
[Abstract]:Objective: to establish a rat model of blood deficiency and liver depression induced by irradiation combined with restraint stress. Methods: male SD rats were randomly divided into blank group, irradiation group, bound group, model group, model group and model group. The body mass, peripheral blood and sugar water, open box test and jumper test were measured in each group. The contents of adrenocorticotropic hormone (ACTH), cortisone, hypothalamic corticotropin releasing hormone (CRH) and hippocampal monoamine neurotransmitter 5-hydroxytryptamine (5-HT) and dopamine (DA) were measured. The contents of peripheral white blood cells, erythrocytes and hemoglobin decreased significantly (P0.05), the contents of P0.05Cort and CRH in hypothalamus increased significantly in plasma, and the contents of monoamine neurotransmitters 5-HT and DA in brain decreased P0.050.Conclusion: from hemogram, behaviorology, the contents of monoamine neurotransmitters 5-HT and DA are significantly decreased. The hypothalamus-pituitary-adrenal gland (HPA) axis and monoamine neurotransmitters in the brain proved that the rat model induced by irradiation combined with restraint stress could be used as an animal model of blood deficiency and liver depression in traditional Chinese medicine.
