大腸桿菌表達的病毒樣顆粒疫苗

【摘要】:重組病毒樣顆粒是病毒衣殼蛋白外源表達的重要形式,形態(tài)結(jié)構(gòu)與天然病毒高度相似,位于納米尺度的大小易于被免疫系統(tǒng)識別,可激發(fā)機體產(chǎn)生保護性免疫反應(yīng),且不含有病毒基因,因此,是一種理想的疫苗形式,也是基于結(jié)構(gòu)進行疫苗設(shè)計的重要結(jié)構(gòu)載體。目前已上市的乙型肝炎疫苗、人乳頭瘤病毒疫苗和戊型肝炎疫苗等基因工程疫苗均采用病毒樣顆粒形式。大腸桿菌表達系統(tǒng)被廣泛用于基因工程藥物的生產(chǎn),具有安全性好、生產(chǎn)周期短、易于放大生產(chǎn)等優(yōu)點,在病毒樣顆粒疫苗應(yīng)用上具有良好前景。本文綜述了利用大腸桿菌研制戊型肝炎疫苗和人乳頭瘤病毒疫苗的進展,特別是這些病毒樣顆粒疫苗的表達及組裝、表位結(jié)構(gòu)特征和臨床試驗結(jié)果。
[Abstract]:Recombinant virus-like particles are an important form of exogenous expression of virus capsid protein. The morphology and structure of recombinant virus particles are similar to those of natural viruses. The size of recombinant virus particles at nanometer scale is easy to be recognized by the immune system, which can stimulate the body to produce protective immune response. And does not contain the virus gene, therefore, is a kind of ideal vaccine form, also is the important structural vector which carries on the vaccine design based on the structure. The recombinant vaccines such as human papillomavirus vaccine and hepatitis E vaccine are in the form of virus-like particles. Escherichia coli expression system is widely used in the production of genetically engineered drugs with good safety and short production cycle. Because of its advantages such as easy to enlarge production, it has a good prospect in the application of virus-like granular vaccine. This article reviews the development of hepatitis E vaccine and human papillomavirus vaccine using Escherichia coli. In particular, the expression and assembly, epitope structure and clinical trial results of these virus-like granular vaccines.
