復發(fā)性口腔潰瘍家兔模型補充超氧化物岐化酶的作用

【摘要】:背景:復發(fā)性阿弗他潰瘍的具體發(fā)病機制目前醫(yī)學界尚無定論,其中體內(nèi)氧自由基的增多是引起復發(fā)性口腔潰瘍反復發(fā)作的原因之一。目的:觀察超氧化物歧化酶對復發(fā)性阿弗他潰瘍治療效果的影響。方法:在24只復發(fā)性阿弗他潰瘍家兔模型建立后,按隨機數(shù)字表法分為3組。模型對照組作為對比不做處理;陰性對照組家兔每天局部碘甘油擦拭;超氧化物歧化酶組家兔每天進行超氧化物歧化酶生物膠囊溶液灌胃,潰瘍局部用碘甘油進行擦拭。1周后切取口腔潰瘍病變組織進行對比研究。各組家兔造模后,每隔3 d上午同一時間兔耳靜脈采血,流式細胞儀分析T細胞亞群CD3+、CD4+、CD8+以及CD3+/CD8+變化。結果與結論:當潰瘍病變形成后,模型對照組和陰性對照組家兔外周血中細胞CD3+、CD4+、CD4+/CD8+的比值經(jīng)過統(tǒng)計均明顯低于正常,而CD8+細胞數(shù)遠遠大于正常數(shù)值。超氧化物歧化酶組灌服超氧化物歧化酶生物膠囊后,CD3+、CD4+細胞數(shù)開始逐漸升高,而CD8+細胞數(shù)則相反呈下降趨勢。提示應用氧自由基的天然清除劑超氧化物歧化酶就能有效清除潰瘍面中過量的氧自由基,從而達到臨床上治療復發(fā)性阿弗他潰瘍的目的。
[Abstract]:Background: the specific pathogenesis of recurrent aphthous ulcer is still unknown in the medical field. The increase of oxygen free radicals in the body is one of the causes of recurrent recurrent oral ulcer. Objective: to observe the effect of superoxide dismutase on the treatment of recurrent aphthous ulcer. After the establishment of the rabbit model of fatta ulcer, The model control group was not treated as a control group; the negative control group was given daily iodine glycerol swab; and the superoxide dismutase group was given intragastric administration of superoxide dismutase biological capsule solution every day. The lesion tissues of oral ulcer were removed with iodine glycerine for 1 week and the blood was collected from the ear vein at the same time every 3 days in the morning after each rabbit model was made. Flow cytometry was used to analyze the changes of CD4 / CD8 and CD3 / CD8 in T cell subsets. Results and conclusion: the ratio of CD4 / CD8 in peripheral blood of model control group and negative control group was significantly lower than that of normal control group. However, the number of CD8 cells was much larger than the normal value, and the number of CD3 + CD4 cells in the superoxide dismutase group began to increase gradually after the superoxide dismutase group was administered with biological capsules of superoxide dismutase. The number of CD8 cells decreased on the contrary, suggesting that superoxide dismutase (SOD), a natural scavenger of oxygen free radicals, could effectively scavenge excess oxygen free radicals in the ulcer surface, thus achieving the purpose of clinical treatment of recurrent aphthous ulcer.
