興奮韁核對老年大鼠頦舌肌和膈肌肌電的影響

【摘要】: 本實驗?zāi)康脑谟谔接戨姶碳ぜ盎瘜W(xué)刺激韁核(Hb)對老年大鼠呼吸,頦舌肌、膈肌肌電的影響,以進(jìn)一步探明上述核團(tuán)在睡眠呼吸暫停綜合癥(SAS)的發(fā)病中的作用。采用電刺激、電損毀和微量注射的方法。結(jié)果:1.電刺激LHb,可迅速產(chǎn)生類似阻塞性睡眠呼吸暫停綜合癥(OSAS)的呼吸狀態(tài),刺激停止后逐漸恢復(fù)至電刺激前呼吸狀態(tài)。呼吸頻率降低(38%),頦舌肌和膈肌肌電改變表現(xiàn)雙向反應(yīng),先出現(xiàn)瞬間的降低而后反射性升高,并且以升高明顯。頦舌肌肌電積分值下降(22%),然后快速升高至(104%)。膈肌肌電積分值降低(11%),然后迅速反射性升高(23%)。微小電刺激MHb后,呼吸頻率增加(12%)。刺激LHb和MHb對動物的呼吸產(chǎn)生不同的效應(yīng),低強(qiáng)度刺激MHb引起動物呼吸加快,而刺激LHb動物呼吸出現(xiàn)類似OSAS呼吸狀態(tài),呼吸頻率變慢,這可能是不同類型的神經(jīng)元被興奮所致。頦舌肌和膈肌肌電出現(xiàn)先降低而后增加的雙向反應(yīng),尤其以增強(qiáng)效應(yīng)為主,可能是出現(xiàn)類似OSAS狀態(tài)后機(jī)體自身的反饋調(diào)節(jié)所致,其中機(jī)制有待進(jìn)一步的研究。2.電損毀大鼠Hb后呼吸頻率升高(19%),頦舌肌肌電積分值降低(46%),膈肌肌電積分值下降(19%),電損毀韁核后得到的結(jié)果不是很明顯,但是依然有統(tǒng)計意義。3. Hb內(nèi)微量注5-羥色胺(5-HT),呼吸頻率降低(31%),頦舌肌和膈肌肌電改變表現(xiàn)雙向反應(yīng),先出現(xiàn)瞬間的降低而后反射性升高,并且以升高明顯。頦舌肌肌電變化積分值下降(21%),然后快速升高(100%)。膈肌肌電積分值降低(14%)。然后迅速反射性升高(24%)。4.Hb內(nèi)微量注射γ-氨基丁酸(GABA),呼吸頻率增加(26%),頦舌肌肌電積分值下降(33%),膈肌肌電積分值下降(8%)。實驗結(jié)果證明Hb與中縫背核之間有密切的往返纖維聯(lián)系,初步確定5-HT及GABA是Hb影響中縫背核活動的介導(dǎo)物質(zhì)之一,
[Abstract]:The purpose of this study was to investigate the effects of electrical and chemical stimulation of habenular nucleus (HB) on respiration, genioglossus and diaphragmatic myoelectric activity in aged rats, in order to further explore the role of these nuclei in the pathogenesis of sleep apnea syndrome (SAS). Electrical damage and microinjection. Results: 1. Electrical stimulation of LHb can rapidly produce respiratory states similar to obstructive sleep apnea syndrome (OSA). After the stimulation stopped, it gradually returned to the breathing state before electrical stimulation. The respiratory rate was reduced by 38%. The electromyography of genioglossus and diaphragm showed a bidirectional response, with a transient decrease and then a reflex increase. And the increase was obvious. The score of genioglossus decreased 22 times, and then rapidly increased to 104. The integral value of diaphragm decreased 11%, and then the rapid reflex increased 23%. After micro electrical stimulation of MHb, The stimulation of LHb and MHb had different effects on the respiration of animals. Low intensity stimulation of MHb increased the rate of respiration in animals, while stimulating LHb resulted in a similar OSAS respiratory state, and the respiratory rate slowed down. This may be due to the excitation of different types of neurons. The bidirectional response of the genioglossus and the diaphragm muscle to decrease first and then increase, especially the enhancement effect, may be due to the feedback regulation of the body itself after the appearance of a similar OSAS state. Among them, the mechanism needs further study .2.Respiratory frequency increased after HB was damaged by electrolysis. The electromyography score of genioglossus decreased by 46%, the integral value of diaphragmatic muscle decreased by 19%, and the results obtained after electric lesion of habenular nucleus were not very obvious. However, there is still statistical significance .3.The microinjection of 5-hydroxytryptamine into HB reduces the respiratory rate by 31%. The electromyography of genioglossus and diaphragm shows a two-way response, with a transient decrease and then an increase in reflex. The integral value of genioglossus electromyography decreased by 21%, and then increased by 100%. The diaphragmatic myoelectric integral value decreased 14%. Then the rapid reflex increased 24%. 4. Microinjection of GABA A into HB, the respiratory rate increased 26%, and the genioglossus muscle increased 26%, and the muscle of the genioglossus increased by 26%, and the muscle of the genioglossus increased by 26%, and the muscle of the genioglossus increased by 26%. The decrease of electrical integral value and the decrease of diaphragmatic myoelectric integral value show that there is a close fiber connection between HB and the dorsal raphe nucleus, and the results of the experiment show that there is a close connection between HB and the dorsal raphe nucleus. 5-HT and GABA were identified as one of the mediators of HB affecting the activity of dorsal raphe nucleus.
