【摘要】：自1980年北京師范大學(xué)生物系首次用大黃復(fù)制脾虛證動(dòng)物模型以來,已有十多種動(dòng)物模型相繼建立。動(dòng)物模型的建立,使關(guān)于脾虛證較深入的實(shí)驗(yàn)研究成為可能。目前研究者已對(duì)各種脾虛證動(dòng)物在消化吸收、免疫、神經(jīng)、內(nèi)分泌等方面的改變進(jìn)行了探討。但是,實(shí)驗(yàn)研究中常用的脾虛證動(dòng)物模型與臨床脾虛證還存在一定的差距。傳統(tǒng)中醫(yī)認(rèn)為“勞”作為病因包括勞力過度、勞神過度和房勞過度三方面,而現(xiàn)有的“勞倦過度”脾虛證動(dòng)物模型,主要采用力竭運(yùn)動(dòng)造成動(dòng)物身體疲勞,對(duì)動(dòng)物精神的影響不明顯。所謂“勞倦過度”造模方法僅模擬了勞力過度,沒有模擬勞神過度這一因素,與臨床脾虛證有一定差距。另外,多數(shù)脾虛證造模方法為單因素造模,僅有少數(shù)幾種復(fù)合因素脾虛證模型。因此,對(duì)現(xiàn)有“勞倦過度”脾虛證單因素動(dòng)物模型進(jìn)行改善,建立一個(gè)比較接近臨床脾虛證的動(dòng)物模型已成為脾虛證本質(zhì)研究的迫切需求。 為了更好地模擬勞力過度和勞神過度兩方面的致病原因,本實(shí)驗(yàn)在以往力竭游泳單因素塑造脾虛證動(dòng)物模型的基礎(chǔ)上剝奪動(dòng)物的部分睡眠時(shí)間,使動(dòng)物不僅勞力而且勞神。從而建立了一種新的脾虛證動(dòng)物模型——改良型勞倦過度小鼠脾虛證模型。 實(shí)驗(yàn)主要觀察了動(dòng)物的一般體征和消化吸收功能的變化,測(cè)定的指標(biāo)有:D—木糖排泄率、血清淀粉酶活性、胃排空速率、腸推進(jìn)速率、離體小腸張力、胃腸粘膜形態(tài)、血清胃泌素含量、脾臟指數(shù)和胸腺指數(shù)。通過與大黃瀉下法、力竭游泳法塑造的脾虛證動(dòng)物模型比較、藥物反證等幾方面來評(píng)估和驗(yàn)證此方法的成立,并對(duì)其影響動(dòng)物消化系統(tǒng)功能的機(jī)制作了初步探討。 實(shí)驗(yàn)結(jié)果顯示,改良型勞倦過度模型小鼠造模后出現(xiàn)脾虛證的典型外觀表現(xiàn),且客觀指標(biāo)改變與文獻(xiàn)報(bào)導(dǎo)相符,與臨床脾虛證患者變化趨勢(shì)一致。在所選的指標(biāo)中,改良型勞倦過度組小鼠的變化均與臨床脾虛證患者的變化趨勢(shì)一致,且多數(shù)指標(biāo)的變化較大黃組(大黃瀉下法塑造脾虛證動(dòng)物模型)和力竭游泳組(力竭游泳法塑造脾虛證動(dòng)物模型)明顯,而大黃組和力竭游泳組只有部分指標(biāo)的變化與臨床患者一致,因此,改良型勞倦過度法塑造脾虛證動(dòng)物模型優(yōu)于大黃瀉下法和力竭游泳法。運(yùn)用四君子湯治療進(jìn)行反證,小鼠一般體征、各客觀指標(biāo)較自然恢復(fù)組明顯改善,說明改良型勞倦過度法塑造的脾虛證模型符合脾虛證動(dòng)物模
[Abstract]:Since 1980, when the animal model of spleen deficiency syndrome was first replicated with rhubarb in the Department of Biology of Beijing normal University, more than ten animal models have been established one after another. At present, researchers have discussed the changes in digestion, absorption, immunity, nerve, endocrine and other aspects of various spleen deficiency syndrome animals. There is still a certain gap between the animal model of spleen deficiency syndrome and clinical spleen deficiency syndrome. Traditional Chinese medicine believes that "labor" as the etiology includes three aspects: excessive labor, excessive mental exhaustion and excessive room labor. However, the existing animal model of "overfatigue" of spleen deficiency syndrome mainly uses exhausted exercise to cause animal body fatigue, which has no obvious effect on animal spirit. The so-called "overfatigue" modeling method only simulates overwork. There is a certain gap between the model of spleen deficiency syndrome and clinical spleen deficiency syndrome. In addition, most of the methods of making model of spleen deficiency syndrome are single factor model, only a few kinds of multiple factors spleen deficiency syndrome model. It is an urgent need to study the essence of spleen deficiency syndrome by improving the existing single factor animal model of spleen deficiency syndrome and establishing an animal model which is close to clinical spleen deficiency syndrome. In order to better simulate the causes of overwork and overwork, this experiment deprived the animals of part of their sleep time on the basis of the previous exhaustive swimming single factor model of spleen deficiency syndrome. A new animal model of spleen deficiency syndrome, modified model of spleen deficiency in overworked mice, was established. The changes of general signs and digestive and absorption functions of animals were observed. The indexes were: excretion rate of 1: D- xylose, serum amylase activity, gastric emptying rate and intestinal propulsion rate. Small intestinal tension, gastrointestinal mucosal morphology, serum gastrin content, spleen index and thymus index were compared with the animal model of spleen deficiency by using the method of rhubarb diarrhea and exhaustive swimming. The establishment of this method was evaluated and verified in several aspects, and the mechanism of its influence on the digestive system of animals was preliminarily discussed. The results showed that the typical appearance of spleen deficiency syndrome appeared in the modified model mice, and the change of objective indexes was consistent with the literature reports. In the selected indexes, the changes of the modified overfatigue group were consistent with the changes of the clinical spleen deficiency syndrome patients. The changes of most indexes were significant in yellow group (rhubarb diarrhea model) and exhaustive swimming group (exhaustive swimming method for spleen deficiency animal model). The rhubarb group and exhaustive swimming group only part of the changes in the same as the clinical patients, so. The modified model of spleen deficiency syndrome was better than that of rhubarb diarrhea and exhaustive swimming. The general signs and objective indexes of mice were obviously improved compared with the natural recovery group. It shows that the model of spleen deficiency is consistent with the animal model of spleen deficiency.
