【摘要】:胚胎種植(implantation)是一個(gè)復(fù)雜的生理過(guò)程。在此期間胚胎與子宮雙方都處在生長(zhǎng)發(fā)育的動(dòng)態(tài)階段,子宮內(nèi)膜與胚胎相互識(shí)別、彼此相容,有侵入能力的胚胎和具有接受性的子宮內(nèi)膜相互作用,發(fā)生一系列生理生化及形態(tài)改變,即雙方建立起“對(duì)話”機(jī)制,胚胎才能在子宮內(nèi)膜正常植入。在人和哺乳動(dòng)物的正常生殖周期中,子宮內(nèi)膜僅有一個(gè)極短的時(shí)期允許胚胎種植,這段時(shí)期稱“種植窗期”,此時(shí)子宮內(nèi)膜表現(xiàn)出最大的容受性。子宮內(nèi)膜對(duì)胚胎的容受性是胚胎種植成功的關(guān)鍵,如果子宮內(nèi)膜不適合胚胎著床將導(dǎo)致妊娠失敗。 目前國(guó)內(nèi)外有關(guān)子宮內(nèi)膜容受性的研究主要集中在生殖調(diào)控因子和內(nèi)膜超微結(jié)構(gòu)兩方面。生殖調(diào)控因子有多種,如:白血病抑制因子(leukaemia inhibitory factor,LIF)、整合素家族(integrins)、胞飲突(pinopode)、肝素結(jié)合樣表皮生長(zhǎng)因子(heparin-binding epidermal growth factor,HB-EGF)、白細(xì)胞介素Ⅰ的Ⅰ型受體(IL-IRt-I)、粘液素-Ⅰ(MUC-Ⅰ)、一氧化氮合酶(NOS)、雌孕激素受體等。胞飲突的形成是子宮內(nèi)膜超微結(jié)構(gòu)中最具代表性的形態(tài)學(xué)改變。 LIF是一種多生物學(xué)功能的分泌型糖蛋白,為IL-6家族成員,具有廣泛的生物學(xué)活性,最早發(fā)現(xiàn)它具有促進(jìn)小鼠MI型白血病細(xì)胞分化并抑制其增殖而命名。近年來(lái)的研究表明LIF參與了生殖活動(dòng)的各個(gè)環(huán)節(jié),如促進(jìn)卵泡的發(fā)育、胚胎的生長(zhǎng)、分化及著床等,尤其是與胚胎種植關(guān)系密切。Stewart等采用基因重組技術(shù),發(fā)現(xiàn)LIF基因缺陷的雌鼠能夠排卵,其卵子亦能夠受精,但胚泡不能正常植入及發(fā)育,若轉(zhuǎn)移到野生型假孕鼠中,則胚泡能夠正常植入和發(fā)育,提出LIF是動(dòng)物胚泡植入過(guò)程所必需的。隨后,LIF與胚胎的種植關(guān)系引起人們極大的興趣,,成為研究的熱點(diǎn)。人們對(duì)子宮內(nèi)膜表達(dá)LIF的情況進(jìn)行了大量研
[Abstract]:Embryo implantation (implantation) is a complex physiological process. During the embryo and the uterus are both at the dynamic stage of growth and development, mutual recognition, endometrium and embryo compatible invade ability of embryos and has accepted the interaction of endometrium, a series of physiological and biochemical and morphological changes, namely, the two sides established the "dialogue" mechanism, in order to endometrial normal embryo implantation. The normal reproductive cycle in human and mammals, endometrial only for a very short period to allow embryo implantation, this period is called "implantation window", the endometrial receptivity showed the largest. The endometrial receptivity to embryo is the key to the success of embryo implantation, if the endometrium is not suitable for embryo implantation will lead to pregnancy failure.
At present study on endometrial receptivity at home and abroad mainly focus on two aspects of reproductive control factors and endometrial morphology. There are many factors such as the regulation of reproduction: leukemia inhibitory factor (leukaemia inhibitory, factor, LIF), integrin family (integrins), pinopode (pinopode), heparin binding epidermal growth factor (heparin-binding epidermal growth factor, HB-EGF), type I receptor of interleukin 1 (IL-IRt-I), mucin 1 (MUC- 1), nitric oxide synthase (NOS), estrogen and progesterone receptors. Pinopode formation is the morphological changes of endometrial ultrastructure in the most representative.
LIF is a kind of biological function of secreted glycoprotein, is a member of the IL-6 family, has a wide range of biological activity, the earliest found that it can promote the differentiation of mouse MI leukemia cells and inhibit its proliferation and naming. Recent studies have shown that LIF is involved in all aspects of reproductive activities, such as the promotion of follicular development, embryo the growth, differentiation and implantation, especially with embryo implantation is closely related to.Stewart using the gene recombination technique, found that LIF deficient female rats can ovulation, the egg can also be fertilized, but not normal blastocyst implantation and development, if transferred to the wild type of pseudopregnant mice, embryo implantation and development is normal, put forward LIF is required for animal implantation.
