【摘要】：結(jié)核病(Tuberculosis,TB)是由結(jié)核分枝桿菌(Mycobacteriumtuberculosis,MTB)所致的以呼吸系統(tǒng)感染為主的慢性傳染病,是目前對人類健康威脅最大的傳染性疾病�？ń槊�(Bacillus CalmetteGuerin,BCG)是目前預(yù)防 TB 唯一的疫苗,但其保護(hù)效果不穩(wěn)定(保護(hù)率從 0-80%不等)。新的疫苗的研制已刻不容緩。 目的:探討結(jié)核分枝桿菌 H37Ra 在小鼠體內(nèi)定植的情況,以及誘導(dǎo)機(jī)體產(chǎn)生特異性抗結(jié)核細(xì)胞免疫功能的情況。 方法:1.用 H37Ra 和 BCG 免疫 BALB/c 小鼠,分別在第 15、30、60天檢測結(jié)核分枝桿菌在小鼠臟器內(nèi)的定植。2.免疫后第 30、60 天,檢測 BALB/c 小鼠脾淋巴細(xì)胞受到 PPD 刺激后增殖轉(zhuǎn)化的刺激指數(shù)(Stimulation Index,SI)。3. 免疫后第 30、60 天,檢測 BALB/c 小鼠脾淋巴細(xì)胞受到 PPD 刺激后白介素-2(interleukin-2,IL-2)的表達(dá)。4.免疫后第 30、60 天,檢測 BALB/c 小鼠脾淋巴細(xì)胞受到 PPD 刺激后可溶性白介素 2 受體(Solubile interleukin 2 receptor,sIL-2R)的表達(dá)。 結(jié)果:1.H37Ra 和 BCG 均能在 BALB/c 小鼠臟器內(nèi)定植。2.免疫 30、60 天后,H37Ra 組淋巴細(xì)胞刺激指數(shù)分別為 2.81±0.63 和 2.16±0.52,BCG 組刺激指數(shù)分別為 2.23±0.20 和 2.02±0.38,均明顯高于未免疫組(1.33±0.17 和 1.47±0.10),結(jié)果有顯著性差異(P0.05)。3.免疫30、 天后, 60 H37Ra 組脾淋巴細(xì)胞 IL-2 的表達(dá)分別為 126.88±8.11pg/ml和 91.26±7.29pg/ml,BCG 組脾淋巴細(xì)胞 IL-2 的表達(dá)分別為 107.15±
[Abstract]:Tuberculosis tuberculosis is caused by Mycobacterium tuberculosis. MTB) is a chronic infectious disease mainly caused by respiratory infection, which is the most dangerous infectious disease to human health at present. BCG vaccine is Bacillus CalmetteGuerin. BCG is the only vaccine to prevent TB, but its protective effect is unstable (the protection rate ranges from 0 to 80%). The development of new vaccine is urgent. Aim: to investigate the colonization of Mycobacterium tuberculosis H37Ra in mice and the induction of specific anti-tuberculosis cell immune function. Methods BALB/c mice were immunized with H37Ra and BCG respectively at 1530. 60 days after immunization, the colonization of Mycobacterium tuberculosis in mouse viscera was detected. The stimulating index of proliferation and transformation of spleen lymphocytes stimulated by PPD in BALB/c mice was determined. After 60 days, interleukin-2 was detected in splenic lymphocytes of BALB/c mice stimulated by PPD. IL-2) expression. 4. 60th day after immunization.
